目的 观察姜黄素对高糖状态下肾小管上皮细胞中线粒体损伤的影响并探讨其作用机制.方法 将人近端肾小管上皮细胞(HK-2细胞)分为对照组、高糖组、高糖+姜黄素处理组.采用Western Blot检测PINK1、Parkin、LC3Ⅱ、Caspase-3的表达,流式细胞仪检测mtROS的表达.结果 高糖处理的HK-2细胞中PINK1、Parkin、LC3Ⅱ的表达均降低,mtROS及凋亡相关蛋白Caspase-3的表达增加,与高糖组相比,予以姜黄素处理后,PINK1、Parkin、LC3Ⅱ的表达升高,mtROS及Caspase-3的表达降低.结论 姜黄素可能通过调控PINK1/Parkin介导线粒体自噬发挥治疗糖尿病肾病的作用.
Effect of Curcumin on Mitophagy in Renal Tubular Epithelial Cells Treated with High Glucose
Objective To observe the effect of curcumin on mitochondrial damage in renal tubular epithelial cells under high glu-cose,and to explore the action mechanism.Methods Human proximal renal tubular epithelial cells were divided into control group,high-glucose model group,and high-glucose+curcumin-treated group.The expression of PINK 1,Parkin,LC3Ⅱ,and Caspase-3 was determined by Western Blot,and mtROS expression was measured by flow cytometry.Results The expressions of PINK1,Par-kin and LC3Ⅱ were decreased in HK-2 cells treated with high glucose,while the expressions of mtROS and apoptosis-related protein Caspase-3 were increased.Compared with the high glucose group,the expressions of PINK1,Parkin and LC3Ⅱ were increased after curcumin treatment.The expression of mtROS and Caspase-3 was decreased.Conclusion Curcumin may play a role in treating dia-betic kidney disease by regulating pinkl/parkin-mediated mitophagy.
