Objective To observe the effect of curcumin on mitochondrial damage in renal tubular epithelial cells under high glu-cose,and to explore the action mechanism. Methods Human proximal renal tubular epithelial cells were divided into control group,high-glucose model group, and high-glucose+curcumin-treated group. The expression of PINK 1,Parkin,LC3Ⅱ,and Caspase-3 was determined by Western Blot, and mtROS expression was measured by flow cytometry. Results The expressions of PINK1,Par-kin and LC3Ⅱ were decreased in HK-2 cells treated with high glucose, while the expressions of mtROS and apoptosis-related protein Caspase-3 were increased.Compared with the high glucose group, the expressions of PINK1,Parkin and LC3Ⅱ were increased after curcumin treatment. The expression of mtROS and Caspase-3 was decreased. Conclusion Curcumin may play a role in treating dia-betic kidney disease by regulating pinkl/parkin-mediated mitophagy.
