Effect of Curcumin on Mitophagy in Renal Tubular Epithelial Cells Treated with High Glucose
Objective To observe the effect of curcumin on mitochondrial damage in renal tubular epithelial cells under high glu-cose,and to explore the action mechanism.Methods Human proximal renal tubular epithelial cells were divided into control group,high-glucose model group,and high-glucose+curcumin-treated group.The expression of PINK 1,Parkin,LC3Ⅱ,and Caspase-3 was determined by Western Blot,and mtROS expression was measured by flow cytometry.Results The expressions of PINK1,Par-kin and LC3Ⅱ were decreased in HK-2 cells treated with high glucose,while the expressions of mtROS and apoptosis-related protein Caspase-3 were increased.Compared with the high glucose group,the expressions of PINK1,Parkin and LC3Ⅱ were increased after curcumin treatment.The expression of mtROS and Caspase-3 was decreased.Conclusion Curcumin may play a role in treating dia-betic kidney disease by regulating pinkl/parkin-mediated mitophagy.
