Objective The purpose of this study was to investigate the expression and mechanism of GEO in the database,tissue samples and breast cancer cells.Methods The expression of hsa_circRNA_048764 in GSE165884 was analyzed by bioinformatics meth-ods,and the expression of hsa_circRNA_048764 in breast cancer tissues and cell lines was detected by real-time quantitative PCR.CCK-8 detects the effect of hsa_circRNA_048764 on the proliferation of breast cancer cell lines.Bioinformatics techniques were used to pre-dict downstream genes of hsa_circRNA_048764.Real-time quantitative PCR and Western Blot were used to detect downstream gene ex-pression.Bioinformatics was used to analyze the effect of hsa-miR-96-5p on the prognosis of breast cancer,and the relationship be-tween the X-box binding protein 1(XBP1)and immune invasion of breast cancer,as well as the effect on the prognosis of immunother-apy.Results Hsa_circRNA_048764 was highly expressed in breast cancer tissues and breast cancer cells in GSE165884 database.Knockdown of hsa_circRNA_048764 expression inhibited the proliferation of breast cancer cells and promoted the expression of hsa-miR-96-5p in breast cancer cells.Overexpression of hsa-miR-96-5p inhibited the expression of XBP1 protein.XBP1 is closely related to CD8+T cells.XBP1 is associated with prognosis in patients treated with anti-PD-L1 immunotherapy.Conclusion Hsa_circRNA_048764 was highly expressed both in breast cancer tissues and cell lines.Knockdown of hsa_circRNA_048764 could inhibit the prolifera-tion of breast cancer cells,and its molecular mechanism may be that it competitively binds the expression of hsa-miR-96-5p and regu-lates the expression of XBP1 protein.XBP1 is involved in the immune microenvironment of breast cancer and is associated with anti-PD-L1 immunotherapy.
