响应面法优化碱蓬蛋白质提取工艺及抗氧化活性分析

目的 对碱蓬蛋白质的提取工艺进行优化,并对其抗氧化活性进行研究,为碱蓬蛋白质的开发及利用提供理论基础.方法 选取碱提酸沉法提取碱蓬中的蛋白质,以碱蓬蛋白质提取率为指标,考察液料比、时间、温度和pH值对碱蓬蛋白质提取率的影响.采用Box-Behnken响应面法进行实验设计,优化碱蓬蛋白质提取工艺.结果 通过响应面分析得到碱蓬蛋白质的提取率为9.73%,抗氧化活性实验得到碱蓬蛋白质对DPPH自由基(DPPH·)和羟基自由基(·OH)的最高清除率分别为83.20%、51.30%.结论 碱蓬蛋白质优化提取工艺具有可行性;抗氧化活性实验证明碱蓬蛋白质具有较强的抗氧化性.
Analysis on Optimization of Extraction Technology of Suaeda salsa Protein by Response Surface Methodology and Its Antioxidant Activity
Objective To optimize the extraction process of Suaeda salsa protein and study its antioxidant properties,providing a theoretical basis for the development and utilization of Suaeda salsa protein.Methods The alkaline extraction and acid precipitation method was selected to extract protein from Suaeda salsa.The extraction rate of Suaeda salsa protein was used as an indicator to investigate the effects of liquid to material ratio,time,temperature,and pH value on protein extraction rate.The Box-Behnken response surface methodology was used to optimize the protein extraction process of Suaeda salsa.Results The extraction rate of Suaeda salsa protein obtained through response surface methodology was 9.73%.The antioxidant activity experiment showed that Suaeda salsa protein had the highest clearance rates for DPPH radcal(DPPH·)and hydroxyl radcal(·OH),which were 83.20%and 51.30%,respectively.Conclusion The optimized extraction process of Suaeda salsa protein is feasible;the antioxidant activity experiment has proved that Suaeda salsa protein has strong antioxidant properties.
