植物病原细菌基因敲除技术在本科开放实验中的设计与实践

基因敲除技术在植物病原细菌和寄主互作研究中发挥着重要作用。近年来,该技术在植物病理学研究领域得到了广泛应用。文章以植物病原细菌基因敲除技术为教学内容,以构建野油菜黄单胞菌(Xanthomonas campestris pv。campestris)的RNA聚合酶σ54因子编码基因rpoN2的突变体为例,探讨了基因敲除技术在本科开放实验教学中的设计与实践。通过rpoN2基因敲除载体构建、野油菜黄单胞菌的遗传转化以及菌落PCR筛选重组子等多方面的教学内容的设计与实践,提高了学生对植物病原细菌基因敲除原理的理解和掌握,培养了学生相关实验操作技能、数据分析和解读能力,以及团队合作精神和科学探究意识。
Design and Practice of Gene Knockout Technology of Plant Pathogenic Bacteria in Undergraduate Open Experiment
Gene knockout technology plays an important role in studying the interaction between pathogenic bacteria and plants.In recent years,this technology has been widely applied in the field of plant pathology.This article takes gene knockout technology in plant pathogenic bacteria as the teaching content and uses the construction of Xanthomonas campestris pv.campestris RNA polymerase σ54 factor coding gene rpoN2 mutant as an example to explore the design and practice of gene knockout technology in undergraduate open experiments.Through various teaching contents such as constructing rpoN2 gene knockout vectors,genetic transformation of X.campestris pv.campestris,and screening recombinants by colony PCR,the aim is to enhance students'understanding and mastery of the principles of gene knockout in plant pathogenic bacteria,cultivate their scientific exploration consciousness,experimental operation skills,data analysis and interpretation abilities,promote their teamwork spirit,and improve their scientific research capabilities.
