Design and Practice of Gene Knockout Technology of Plant Pathogenic Bacteria in Undergraduate Open Experiment
Gene knockout technology plays an important role in studying the interaction between pathogenic bacteria and plants.In recent years,this technology has been widely applied in the field of plant pathology.This article takes gene knockout technology in plant pathogenic bacteria as the teaching content and uses the construction of Xanthomonas campestris pv.campestris RNA polymerase σ54 factor coding gene rpoN2 mutant as an example to explore the design and practice of gene knockout technology in undergraduate open experiments.Through various teaching contents such as constructing rpoN2 gene knockout vectors,genetic transformation of X.campestris pv.campestris,and screening recombinants by colony PCR,the aim is to enhance students'understanding and mastery of the principles of gene knockout in plant pathogenic bacteria,cultivate their scientific exploration consciousness,experimental operation skills,data analysis and interpretation abilities,promote their teamwork spirit,and improve their scientific research capabilities.
