摘要

采用静态生物测定方法,将三疣梭子蟹暴露培养在含有不同Cu2+浓度的海水中(盐度22±1,pH7.5±0.5,温度(18±1)℃),分别在24、48、72和96h对三疣梭子蟹进行死亡个数统计,通过机率单位法构建三疣梭子蟹24、48、72和96 h浓度对数与机率单位的直线蚓归方程,并根据回归方程计算出三疣梭子蟹24、48、72和96 h的半数致死浓度(LC5D)分别为51.20、22.93、19.74和9.74 mg/L.在此基础上,采用湿法灰化法测定了在Cu2+浓度5、13.5、18和24 mg/L下暴露7 d后,三疣梭子蟹鳃、肌肉、肝胰腺三种组织中Cu2+的生物积累量.结果表明:三种组织中Cu2的积累量差异较大,并且随着水中Cu2+浓度的增高而逐渐增高;三种组织中鳃积累最快也最高,肝胰腺次之,肌肉最低.说明Cu2+的积累与机体组织的结构和功能有关.

Abstract

Static bioassay tests were employed in this experiment. Portunus trituberculatus were exposed in the sea water (21±1‰,pH 7.5±0.5,18±1 ℃) including different concentrations of copper (13.5,18,24,32,56,75 rog/L) for 24 h,48h. 72 h,96 h. The number of the dead P. trituberculatus was counted to determine the median lethal concentration(LC50)of 24 h,48 h,72 h,96 h; four linear regression equations of 24 h,48 h. 72 h,96 h were established which expressed the relationship between the probit and concentration logarithm with the probit method,according to the four equations,the LC50 of 24、48、72 and 96 h were calculated and were 51.20、22.93、19.74 and 9.74 mg/L,respectively. Based on the above,the bioaccumulation of Cu2+ in the three main tissues of P. trituberculatus were measured through the wet-ash method with exposing crabs for 7d under 5,13.5,18,24 mg/L. The result demonstrated that there was a obvious difference amongst the bioaccumulation of Cu2+ in three tissues,and a distinct increase of which were observed with the increasing concentrations of Cu2+; the bioaccumulation of Cu2+ in gills were the highest and grow the fastest,followed by the hepatopancreas and muscle.
