越南人参应答Neofusicoccum ribis侵染的生理指标检测及N.ribis生理特性的鉴定

越南人参(Panax vietnamnensis Ha & Grushv.)是一种具有高附加值的药用植物,在人工栽培过程中极易受到病害的影响,叶枯病即是越南人参最主要的病害之一,其病原真菌为Neofusicoccum ribis(N.ribis).通过病原真菌侵染实验评价了 N.ribis的致病力,在接种后2 d就能出现病症,证实N.ribis对越南人参具有很强的侵染能力.对患病后越南人参生理指标进行分析,发现叶绿素随着真菌侵染时间的增加逐渐下降,其体内的过氧化物酶、超氧化物歧化酶、过氧化氢酶和活性氧含量会在前4 d急剧增加,并在第4 d达到峰值.对N.ribis的生物学特性进行鉴定和分析,发现其最适生长培养基为马铃薯葡萄糖琼脂培养基、最适碳源为果糖、最适氮源为硝酸钠、最适温度为25~30 ℃、最适pH为4.0~7.0,光照有利于其生长.研究结果将为越南人参叶枯病的防控提供理论指导.
Detection of Physiological Indicators of Panax vietnamensis Ha &Grushv.in Response to Neofusicoccum ribis Infection and Identification of the Physiological Characteristics of N.ribis
Panax vietnamensis Ha&Grushv.is a medicinal plant with high added value in Yunnan Province,which is easily affected by diseases in the process of cultivation.Leaf blight is one of the most dangerous disease of P.vietnamensis,and its pathogen has been identified as Neofusicoccum ribis(N.ribis).In this study,the patho-genicity of N.ribis was evaluated by fungal inoculation experiment.Two days after inoculation,the symptoms of P.vietnamensis began to occur,which confirmed that N.ribis had strong infection ability to P.vietnamensis.The physiological indexes of P.vietnamensis after suffering from leaf blight were analyzed,the results showed that chlo-rophy gradually decreased with the increase of the N.ribis infection time,the contents of peroxidase,superoxide dismutase,catalase and reactive oxygen species in P.vietnamensis increased sharply in four days after inoculation and reached the peak on the fourth day.In addition,the biological characteristics of N.ribis were identified and analyzed,and it was found that the optimum growth medium was PDA,the optimum carbon source was fructose,the optimum nitrogen source was sodium nitrate,the optimum temperature was 25~30 ℃,the light was beneficial to its growth,and the optimum pH was 4.0~7.0.The results will lay a foundation and provide research ideas for the prevention and control of leaf blight in P.vietnamensis.
