目的:探究hsa_circ_0030042 在口腔鳞状细胞癌(OSCC)中的作用.方法:通过实时荧光定量PCR检测OSCC细胞中hsa_circ_0030042 的表达,在OSCC细胞中转染小干扰RNA(siRNA)以敲低hsa_circ_0030042,采用CCK-8、集落形成实验、划痕实验、Transwell实验检测hsa_circ_0030042 对OSCC细胞的增殖、侵袭、迁移作用影响.结果:hsa_circ_0030042 在CAL27 和HSC3 细胞中的表达显著升高(P<0.001);与对照组相比,敲低hsa_circ_0030042 后,CAL27、HSC3 细胞的增殖、迁移和侵袭能力均显著降低(P<0.05).结论:hsa_circ_0030042 在OSCC细胞中的表达显著增高,可促进OSCC细胞的增殖、侵袭和迁移.
Study on the role of hsa_circ_0030042 in oral squamous cell carcinoma
Objective:To identify the role of hsa_circ_0030042 in oral squamous cell carcinoma(OSCC).Methods:The expres-sion of hsa_circ_0030042 in OSCC cells was detected by qRT-PCR.Transfection of small interfering RNA(siRNA)was performed in OSCC cells to knock down hsa_circ_0030042.The effect of hsa_circ_0030042 on OSCC was validated using CCK-8 assay,colony for-mation assay,cell scratch assay and Transwell assay.Results:The expression of hsa_circ_0030042 in CAL27 and HSC3 cells was sig-nificantly increased(P<0.001).Compared with the control group,the proliferation of CAL27 and HSC3 cells was significantly de-creased after knockdown of hsa_circ_0030042(P<0.05).The migration and invasion ability of CAL27 and HSC3 cells were both sig-nificantly decreased(P<0.05).Conclusions:Hsa_circ_0030042 is significantly upregulated in OSCC and can promote proliferation,invasion and migration in OSCC.
