Study on the role of eRNA ENSR00000313345 in the tumorigenesis and development of head and neck squamous cell carcinoma
Objective:To identify the role of eRNA ENSR00000313345 in head and neck squamous cell carcinoma(HNSCC).Methods:Co-expression between individual survival-related eRNA ENSR00000313345 and its putative target gene WWC2 was identi-fied by Spearman's correlation analysis.This expression of eRNA ENSR00000313345 in HNSCC cell lines was detected by qRT-PCR technology.Small interfering RNA was transfected into HNSCC cell lines to knock down eRNA ENSR00000313345,followed by func-tional experiments including CCK-8 assay,cell scratch assay,and Transwell migration assay to verify the effect of eRNA EN-SR00000313345 on HNSCC.Small interfering RNA was transfected into HNSCC cell lines to knock down WWC2,and CCK-8 assay,cell scratch assay,and Transwell migration assay were performed to detect the effect of WWC2 on HNSCC.The correlation between eR-NA ENSR00000313345 and WWC2 was examined using qRT-PCR and Western blot techniques.Results:The expression level of WWC2 was positively correlated with eRNA ENSR00000313345.eRNA ENSR00000313345 exhibited higher expression levels in HNSCC cell lines compared to oral mucosal keratinocyte cell lines(P<0.05).Knocking down eRNA ENSR00000313345 suppressed the proliferation and migration capabilities of HNSCC cell lines(P<0.05).Knocking down WWC2 inhibited the proliferation and mi-gration abilities of HNSCC cell lines(P<0.05).The expression levels of WWC2 decreased concomitantly with the knockdown of eRNA ENSR00000313345(P<0.05).Conclusions:eRNA ENSR00000313345 can promote the proliferation and migration of HNSCC cells by regulating WWC2 expression.
