基于代谢组学研究XLS对白色念珠菌的代谢变化及抑菌机制

目的:血链球菌提取物XLS是从口腔血链球菌中提取的具有抑菌作用的胞内蛋白,本文通过非靶向代谢组学技术检测分析XLS作用于白色念珠菌所产生的代谢变化,并初步分析其作用机制.方法:(1)非靶向代谢组样本制备:实验组:0.5 mg/mL XLS与白色念珠菌共培养12 h对照组:单纯白色念珠菌培养12 h,每组6个生物学重复.(2)利用非靶向代谢组学技术对两组样本进行代谢变化研究,运用多变量分析[主成分分析(principal compo-nents analysis,PCA)和偏最小二乘法判别分析(partial least squares discriminant analysis,PLS-DA)]和单变量分析(t检验)比较两组样本之间的差异.结果:PLS-DA和PCA分析结果显示两组样本差异性显著.本研究筛选出160个正离子差异代谢物,其中68个代谢物上调,92个代谢物下调;90个负离子差异代谢物,其中52个代谢物上调,38个代谢物下调.通过京都基因与基因组百科全书(Kyoto Encyclopedia of Genes and Genomes,KEGG)富集分析发现,富集显著的通路主要包括抗坏血酸与醛酸代谢通路、氨基酸合成、嘌呤嘧啶代谢通路.此外,还发现了一些与氧化应激反应相关的代谢物,与对照组相比,其在实验组均表达上调.结论:(1)XLS对白色念珠菌的氨基酸代谢、脂质代谢、嘌呤嘧啶代谢等过程产生显著影响;(2)XLS可能通过引起白色念珠菌菌体的氧化应激反应导致白色念珠菌细胞的氧化损伤,从而抑制白色念珠菌的生长繁殖.
The Metabolic Changes and Inhibitory Mechanism of XLS Against Candida Albicans were Studied Based on Metabolo-mics
Objective:Streptococcus sanguis extract XLS is a bacteriostatic intracellular protein extracted from oral Streptococcus sanguis.To detect and analyze the metabolic changes of XLS on Candida albicans by non-targeted metabolomics,and to analyze its mechanism.Methods:The samples were prepared firstly.Experimental group:0.5 mg/mL XLS was co-cultured with Candida albicans for 12 h;control group:pure Candida albicans was cultured for 12 h,with 6 biological replicates in each group.Two non-targeted metabolomics technologies were used to study the metabolic changes of samples.Multivariate analysis[(principal components analysis,PCA)and(partial least squares discriminant analysis,PLS-DA)]and univariate analysis(t test)were used to compare the differences.Re-sults:PLS-DA and PCA analysis showed significant differences between two groups of samples.In this study,160 positive ion differential metabolites were screened,of which 68 metabolites were up-regulated and 92 metabolites were down-regulated.Among the 90 negative ion differential metabolites,52 metabolites were up-regulated and 38 metabolites were down-regulated.Through KEGG enrichment analysis,it was found that the pathways with signifi-cant pathway enrichment mainly included ascorbic acid and aldehyde acid metabolic pathways,amino acid synthesis,purine pyrimidine metabolic pathways,etc.In addition,some metabolites related to oxidative stress were also found,which were up-regulated in the experimental group compared with the control group.Conclusion:XLS has a significant effect on amino acid metabolism,lipid metabolism,purine and pyrimidine metabolism,and other proces-ses of Candida albicans.XLS causes oxidative damage to Candida albicans cells by causing oxidative stress in Candi-da albicans,thereby inhibiting the growth and reproduction of Candida albicans.
