摘要

采用生物信息学方法挖掘皮肌炎相关的铁死亡基因,为皮肌炎的临床诊断及治疗提供新思路.从基因表达综合数据库中筛选出关于皮肌炎的数据集,应用R语言对数据集进行分析,得到皮肌炎的261个差异基因并与FerrDb数据库中的铁死亡基因取交集后得到15个皮肌炎相关的铁死亡基因,使用基因本体富集,京都基因和基因组百科全书途径显示它的作用可能和炎症、铁死亡、氧化应激和凋亡等相关,基于蛋白质-蛋白质相互作用网络分析及Cytoscape里的算法筛选出5个关键基因皮肌炎相关的铁死亡基因,通过基因表达和受体工作特征曲线进一步确定了 4个诊断基因TIMP1、HMOX1、TFRC、CDK1A作为皮肌炎诊断及治疗的生物标志物的潜力,为皮肌炎的铁死亡机制提供新的见解.

Abstract

The ferroptosis genes associated with dermatomyositis are mined by bioinformatics to provide a new idea for clinical diagnosis and treatment of dermatomyositis.The dataset about dermatomyositis was screened from the comprehensive gene expression database,and the dataset was analyzed using R language.261 differential genes of dermatomyositis were obtained,and 15 ferroptosis genes related to dermatomyositis were obtained after intersection with ferroptosis genes in FerrDb database.According to the Kyoto Encyclopedia of Genes and Genomes pathway,its role may be related to inflammation,ferroptosis,oxidative stress,and apoptosis.Based on protein-protein interaction network analysis and Cytoscape selected five key genes associated with dermatitis and ferroptosis.The potential of four diagnostic genes TIMP1,HMOX1,TFRC and CDK1A as biomarkers for the diagnosis and treatment of dermatomyositis is further identified through gene expression and receptor working characteristic curves,providing new insights into the mechanism of ferroptosis in dermatomyositis.
