On the causal relationship between gut microbiota and allergic rhinitis based on Mendelian randomization
Objective To analyze the causal relationship between gut microbiota and allergic rhinitis based on Mendelian randomization.Methods The human gut microbiota dataset was downloaded from the MiBioGen database,and 196 known bacterial groups(9 phyla,16 classes,20 orders,32 families and 119 genera)were retained as exposure factors.Single nucleotide polymorphisms that were strongly correlated with exposure factors and independent of each other were selected as effective instrumental variables.An allergic rhinitis dataset(finn-b-ALLERG_RHINITIS)with 5 527 patients and 212 387 controls was downloaded from the genome-wide association studies database as the outcome variable.A two-sample Mendelian randomization analysis was performed using inverse variance weighted,weighted median,MR-Egger,weighted model and simple model methods,respectively.The causal association between gut microbiota and allergic rhinitis was evaluated by odds ratio(OR).Sensitivity analysis was performed by leave-one-out method.Horizontal pleiot-ropy was tested by MR-Egger intercept test and MR-PRESSO global test,and Cochran's Q test used for heterogeneity.Results Inverse variance weighted analysis showed that Victivallaceae(OR=1.111,95%CI:[1.004,1.230],P=0.042)and Ruminococcus gauvreauii group(OR=1.263,95%CI:[1.044,1.528],P=0.016)were significantly positively correlated with allergic rhinitis,while Ruminococcaceae UCG-010(OR=0.775,95%CI:[0.605,0.993],P=0.044),Subdoligranulum(OR=0.735,95%CI:[0.595,0.909],P=0.004),Burkholderiales(OR=0.764,95%CI:[0.606,0.963],P=0.023)and Actinobacteria(OR=0.798,95%CI:[0.657,0.970],P=0.024)negatively correlated with it.In addition,the results of leave-one-out sensitivity analysis were stable,MR-Egger intercept test and MR-PRESSO global test showed no horizontal pleiotropy,and Cochran's Q test showed no heterogeneity.Conclusion A causal relationship existed between gut mi-crobiota and allergic rhinitis.Victivallaceae and Ruminococcus gauvreauii group might increase the risk of allergic rhinitis.Ruminococcaceae UCG-010,Subdoligranulum,Burkholderiales and Actinobacteria could reduce its risk.
