Using high-resolution liquid chromatography tandem mass spectrometry method to analyze the triglyceride profile in M1 and M2 macrophages
Objective To analyzed the lipid profiles of M1 and M2 macrophages via high-resolution liquid chromatography tandem mass spectrometry(LC-MS/MS)so as to reveal the important role played by lipid metabolism,especially triglyceride,in macrophage polarization.Methods Polarized M1 and M2 macrophages were generated from bone marrow-derived macrophages,and assessed the expression levels of marker genes in polarized macrophages by quantitative polymerase chain reaction,high-resolution LC-MS/MS was used to analyze the triglyceride profile of polarized macrophages and determine the difference between polarized macrophages.Results 61 triglyceride species were detected in polarized macrophages.The total triglycer-ide level was upregulated in M1 macrophages,but downregulated in M2 macrophages.Most of the detected triglycerides contain long-chain fatty acids.Triglycerides with long-chain polyunsaturated fatty acids were upregulated in M1 macrophages,but downregulated in M2 macrophages.Conclusion Macrophage polariza-tion lead to a significant change of the triglyceride profile.
