Application of DGAT1 as a marker for clinical diagnosis of parotid adenolymphomas
Objective To investigate the expression and clinical significance of diglycerol O-acyltransferase 1(DGAT1)in parotid adenolymphomas tissue.Methods Clinical data of adenolymphoma patients from Shandong University Qilu Hospital Dezhou Hospital from January 2012 to December 2021 were collected,and their age,gender,unilateral and bilateral onset,smoking,tumor location and size,and blood lipids of the patients analyzed.Samples of parotid adenolymphomas,pleomorphic adenoma,and normal parotid tissue,each from 5 cases,were selected.The expression of DGAT1 in each tissue was detected by reverse tran-scription quantitative polymerase chain reaction(RT-qPCR)and Western blotting.Immunohistochemical staining was performed on pathological sections of 30 patients to detect the expression differences of DGAT1 in parotid adenolymphomas and normal parotid tissue.Results The ages of 111 patients with parotid ade-nolymphomas ranged from 25 to 84 years,with an average age of(62.08±10.61)years;92 were male(82.9%);79 had a smoking history(71.2%),with smoking duration ranging from 8 to 50 years;103 had unilateral disease(92.8%);62 had tumors located at the tail of the parotid gland(55.9%);tumor diameters ranged from 0.5 to 6.2 cm,with an average diameter of(2.998±1.083)cm;76 had tumors with diameters ranging from 2.0 to 4.0 cm(68.5%).RT-qPCR and Western blotting results showed that the expression of DGAT1 in parotid adenolymphomas was significantly higher than that in pleomorphic adenoma and nor-mal parotid tissue.Immunohistochemical results showed a high expression of DGAT1 in parotid adenolym-phomas.Conclusion Adenoma is more common in middle-aged and elderly male smokers.The lesions are mostly located in the posterior lower pole of the parotid gland,and the tumor size is mostly 2.0~4.0 cm.DGAT1 is highly expressed in parotid adenolymphomas,significantly higher than in normal glandular tissue,indicating a close correlation between a high expression of DGAT1 and the occurrence of parotid adenolym-phomas,which is expected to become a new biomarker for the diagnosis of adenolymphoma.
