Constructing a disulfidptosis-related lncRNA model to predict the prognosis and immune related analysis of gastric cancer patients
Objective To construct a gastric cancer prognosis model based on disulfidptosis-related long non-coding RNA(lncRNA),analyze the association between lncRNAs and the tumor immune microenvironment of gastric cancer,and evaluate potential immunotherapy.Methods RNA sequencing data and clinical data of TCGA-STAD were downloaded,an lncRNA risk prognostic model associated with disulfidptosis established using coexpression network analysis,Cox regression analysis and LASSO regression.In addition,receiver operating characteristic curves and other methods were used to verify the predictive ability of the model.Then,gene set enrichment analysiswas used to analyze differential pathways between the two groups.The relation-ship between tumor microenvironment,immune cells and immune function was evaluated.Results Based on previous literature,24 disulfidptosis related genes and 9 disulfidptosis related lncRNAs were collected for con-structing prognostic models.Patients were divided into high-risk and low-risk groups,and the overall survival rate between the two was statistically significant(P<0.001)from the survival curve.The area under the curve of the prediction models at 1 year,3 years and 5 years were 0.681,0.703 and 0.789,respectively,indicating a strong predictive ability.Cox regression analyses both proved that the risk score to be an independent factor af-fecting prognosis(P<0.001).There were significant differences in some immune cells and immune function between high and low risk groups(P<0.05).Conclusion The risk score model based on 9 lncrnas associated with disulfidptosis is helpful to predict the prognosis and immune response of patients with gastric cancer,and can provide a reference for individualized treatment of patients.
