Physicochemical characteristics of graded alcohol precipitation Herba Patriniapolisa Calid and inhibition of sugar-digesting enzyme activity
Objective To investigate the physicochemical characteristics and glycodigestive enzyme inhibitory activity of different fractions of Herba patriniae polysaccharides,as well as the correlation between them.Methods Four kinds of HPs(HP-50,HP-60,HP-70 and HP-80)were obtained by hot water extraction and alcohol precipitation at different concentrations of ethanol(50%,60%,70%,80%),and the content and mono-saccharide composition of polysaccharides determined by phenol-sulfuric acid method,gas chromatography(GC),high performance liquid chromatography(GPC)and ultraviolet spectrophotometry(UV).The activities of four polysaccharides in inhibiting sugar-digesting enzymes were compared via a bivariate correlation analy-sis regarding their physical and chemical characteristics such as molecular weight and inhibitory activity against α-amylase and α-glucosidase.Results All HPs exhibited concentration-dependent inhibition of sugar-digesting enzymes,and HP-80 had the best inhibitory activity of sugar-digesting enzymes.Compared with oth-er alcohol precipitation concentrations,HP-80 had a higher glucose content,rhamnose and arabinose content was lower,and the molecular weight smaller.Conclusion It is speculated that the relative molecular weight of Herba patriniae polysaccharides is 50~100 kDa,the molar fraction of rhamnose less than 9.83%and the amount ratio of arabinose to glucose substance less than 1,which could more conducive to the inhibition of α-amylase and α-glucoside activities of HPs.
