Objective To investigate the expression of WIF-1 and its relationship with serum inflammatory factors,Atrogin-1/MuRF-1 protein levels in chronic obstructive pulmonary disease(COPD)combined with skeletal muscle atrophy rats.Methods Thirty SD rats were di-vided into the COPD group(rats with successful COPD model establishment,n=11),the COPD combined with skeletal muscle atrophy group(rats with COPD combined with malnutrition-induced skeletal muscle atrophy,n=9)and the control group(healthy rats without animal model establishment,n=10)according to the random number table method,and treated accordingly.The levels of serum interleukin(IL)-6,IL-8,and tumor necrosis factor(TNF)-α in the three groups of rats were detected by enzyme-linked immunosorbent assay;lung tissue and gastrocne-mius muscle tissue morphology was observed by Hematoxylin-Eosin staining;the cell-positive expression rate of WIF-1 in skeletal muscle tis-sue was detected by immunohistochemistry;the protein expression of Atrogin-1 and MuRF-1 in skeletal muscle tissue was detected by Western blotting;the correlation between WIF-1 and Atrogin-1,MuRF-1 was analyzed by Pearson's method.Results Compared with the control group,the levels of IL-6,IL-8,and TNF-α in the COPD group were significantly increased,the differences were statistically significant(P<0.05);compared with the COPD group,the levels of IL-6,IL-8,and TNF-α in the COPD combined with skeletal muscle atrophy group were significantly increased,the differences were statistically significant(P<0.05).Compared with the control group,the cell-positive expres-sion rate of WIF-1 in the COPD group rats was higher,but the difference was not statistically significant(P>0.05);compared with the COPD group,the cell-positive expression rate of WIF-1 in the COPD combined with skeletal muscle atrophy group was significantly increased,the differences were statistically significant(P<0.05).Compared with the control group,the protein levels of Atrogin-1 and MuRF-1 in the COPD group were increased,but the differences were not statistically significant(P>0.05);compared with the COPD group,the protein levels of Atrogin-1 and MuRF-1 in the COPD combined with skeletal muscle atrophy group were significantly increased,the differences were statisti-cally significant(P<0.05).WIF-1 showed a positive correlation with Atrogin-1(r=0.760,P<0.001),but had no correlation with MuRF-1(r=0.149,P=0.433).Conclusion The positive expression of the WIF-1 gene is significantly increased in COPD combined with skeletal muscle atrophy rats,and it plays a role in promoting the inflammatory factor response and can also affect muscle atrophy-related proteins to promote disease progression.
