Objective To investigate the effect of mononoside on the neuroinflammation of rats with Parkinson's disease by regulating the cAMP/PKA signal pathway.Methods Thirty rats were divided into the normal group,the model group and the monoside group according to the random number table method,with 10 rats in each group.The rat models of Parkinsons disease was established by subcutaneous injection of rote-none into the neck and back of rats.Monoside group was given orally monoside 180 mg·kg-1·d-1,equal doses of saline by gavage in normal and model groups.All groups were given gavage once a day for 14 days.The histological changes in hippocampal neurons was observed using he-matoxylin-eosin staining.Rats in each group were intraperitoneally injected with 0.5 mg/kg dehydrated morphine hydrochloride to induce rats to choose to the left,and the rotation duration and rotation frequency were recorded;the levels of interleukin(IL)-1β,IL-6,tumor necrosis factor-α(TNF-α),cAMP and PKA in brain tissue were determined by enzyme-linked immunosorbent assay.The expression of cAMP/PKA mRNA was determined by real-time fluorescence quantitative PCR;the expression of cAMP/PKA protein was determined by Western blotting.Results The rotation frequency and duration of rats in the model group and the mononoside group were higher(longer)than those in the normal group,therotation frequency and duration of rats in the mononoside group were lower(shorter)than those in the model group,the differences were statistically significant(P<0.05).The levels of IL-1 β,IL-6 and TNF-α in the model group and the mononoside group were higher than those in the normal group,the levels of IL-1 β,IL-6 and TNF-α in the monoside group were lower than those in the model group,the differ-ences were statistically significant(P<0.05).The levels of cAMP and PKA of rats in the model group and the mononoside group were lower than those in the normal group,the levels of cAMP and PKA of rats in the mononoside grouphigher than those in the model group,the differences were statistically significant(P<0.05).The relative expression of cAMP and PKA mRNA of rats in the model group and the mononoside group were lower than those in the normal group,the relative expression of cAMP and PKA mRNA of rats in the mononoside group were higher than those in the model group,the differences were statistically significant(P<0.05).The gray values of cAMP and PKA protein of rats in the model group and the mononoside group were lower than those in the normal group,the gray values of cAMP and PKA protein of rats in the mononoside group were higher than those in the model group,the differences were statistically significant(P<0.05).Conclusion Monoside can reduce neuroin-flammation in rats with Parkinson's disease,and its mechanism may be related to the activation of cAMP/PKA signal pathway.
