摘要
目的 结直肠癌患者血清微小RNA(miR)-150-5p和miR-200b-3p水平及临床意义.方法 回顾性选择2020年6月至2022年11月在沧州市人民医院就诊的结直肠癌患者116例为结直肠癌组,同时选取同期在本院治疗的结肠息肉患者60例作为结直肠良性肿瘤组,选择同期在本院行健康体检者60名为对照组.采用实时荧光定量PCR(qRT-PCR)检测受试者血清miR-150-5p、miR-200b-3p水平;采用Pearson相关性分析对结直肠癌患者血清miR-150-5p与miR-200b-3p表达水平的相关性进行分析;分析结直肠癌患者血清miR-150-5p、miR-200b-3p水平与病理特征的关系;采用受试者操作特征(ROC)曲线分析血清miR-150-5p、miR-200b-3p表达水平对结直肠癌的诊断价值.结果 结直肠癌组、结直肠良性肿瘤组血清miR-150-5p、miR-200b-3p水平均显著低于对照组,且结直肠癌组血清miR-150-5p、miR-200b-3p水平均显著低于结直肠良性肿瘤组,差异均有统计学意义(P＜0.05).Pearson相关性分析结果显示,结直肠癌患者血清miR-150-5p与miR-200b-3p表达水平呈正相关(r=0.562,P＜0.05).结直肠癌患者血清miR-150-5p、miR-200b-3p水平与分化程度、淋巴结转移、TNM分期、浸润深度有关(P＜0.05),其中低分化、有淋巴结转移、TNM分期Ⅲ～Ⅳ期、浸润深度T3～T4的结直肠癌患者血清miR-150-5p、miR-200b-3p水平显著低于中高分化、无淋巴结转移、TNM分期Ⅰ～Ⅱ期、浸润深度T1～T2的患者(P＜0.05).血清miR-150-5p、miR-200b-3p单独及联合诊断结直肠癌的曲线下面积(AUC)分别为0.800、0.828、0.893,联合诊断AUC显著高于单独预测AUC(P＜0.05).结论 结直肠癌患者血清miR-150-5p、miR-200b-3p水平降低,二者可作为诊断结直肠癌发生的辅助指标.
Abstract
Objective To investigate the serum levels of microRNA(miR)-150-5p and miR-200b-3p in patients with colorectal cancer and their clinical significance.Methods A total of 116 colorectal cancer patients who admitted to Cangzhou People's Hospital from June 2020 to November 2022 were collected as the colorectal cancer group,meantime,60 patients with colonic polyps treated in our hospital were se-lected as the colorectal benign tumor group and 60 people with healthy physical examination in our hospital were selected as the control group.The serum levels of miR-150-5p and miR-200b-3p in subjects were detected by real-time fluorescence quantitative PCR(qRT-PCR).The correlation between serum miR-150-5p and miR-200b-3p expression levels in colorectal cancer patients was analyzed by Pearson correlation.The relationship between serum miR-150-5p,miR-200b-3p levels and pathological features in patients with colorectal cancer was analyzed.The diagnostic value of serum miR-150-5p and miR-200b-3p expression levels for colorectal cancer was analyzed by receiver operating char-acteristic(ROC)curve.Results The serum levels of miR-150-5p and miR-200b-3p in the colorectal cancer and colorectal benign tumor groups were obviously lower than those in the control group,and the serum levels of miR-150-5p and miR-200b-3p in the colorectal cancer group were obviously lower than those in the colorectal benign tumor group,the differences were statistically significant(P＜0.05).Pearson cor-relation analysis results showed a positive correlation between serum miR-150-5p and miR-200b-3p expression levels in colorectal cancer pa-tients(r=0.562,P＜0.05).The serum levels of miR-150-5p and miR-200b-3p in colorectal cancer patients were related to differentia-tion,lymph node metastasis,TNM staging,and depth of infiltration(P＜0.05),the serum miR-150-5p and miR-200b-3p levels in color-ectal cancer patients with low differentiation,lymph node metastasis,TNM stage Ⅲ-Ⅳ,and infiltration depth T3-T4 were obviously lower than those in patients with medium to high differentiation,no lymph node metastasis,TNM stage Ⅰ-Ⅱ,and infiltration depth T1-T2(P＜0.05).The area under the curve(AUC)of serum miR-150-5p and miR-200b-3p for single and combined diagnosis of colorectal cancer was 0.800,0.828,and 0.893,respectively,the area under the curve of combined diagnosis was obviously higher than the AUC of single prediction(P＜0.05).Conclusion The serum levels of miR-150-5p and miR-200b-3p in colorectal cancer patients decrease,which can serve as auxilia-ry indicators for diagnosing the occurrence of colorectal cancer.
