桂药生精胶囊对少弱精子症模型大鼠caspase-8、caspase-3表达的影响

摘要
目的 观察桂药生精胶囊对少弱精子症模型大鼠睾丸组织胱天蛋白酶(caspase)-8、caspase-3蛋白表达的影响.方法 将40只雄性SD大鼠依据随机数字表法分为空白组(n=10)和造模组(n=30).造模组采用腹腔注射环磷酰胺法构建少弱精子症模型.模型制备成功后,将造模组按照随机数字表法分为模型组、桂药生精胶囊组和阳性对照组,每组各10只.阳性对照组予左卡尼汀口服溶液200 mg/(kg·d)灌胃,桂药生精胶囊组予桂药生精胶囊52.5 mg/(kg·d)灌胃,空白组、模型组灌胃等量0.9%氯化钠溶液,1次/d,持续干预28 d.末次给药24 h后,摘取睾丸及附睾,检测精子质量,光镜下观察睾丸组织形态,采用实时荧光定量聚合酶链式反应和免疫组织化学染色法检测睾丸组织caspase-8、caspase-3的mRNA和蛋白表达.结果 与空白组相比,模型组大鼠精子数量和活力降低,差异有统计学意义(P<0.05);与模型组相比,桂药生精胶囊组、阳性对照组大鼠精子数量和活力均升高,差异均有统计学意义(P<0.05).模型组大鼠较空白组睾丸生精小管萎缩,生精细胞分化程度均显著降低,精子数量明显减少,呈空泡化改变;与模型组相比,桂药生精胶囊组及阳性对照组大鼠睾丸生精小管形态结构得到改善,生精细胞分化程度升高,精子数量显著增多,细胞外膜得以修复.与空白组相比,模型组caspase-8、caspase-3 mRNA表达水平均升高,差异均有统计学意义(P<0.05);与模型组相比,桂药生精胶囊组、阳性对照组caspase-8、caspase-3 mRNA表达水平均降低,差异均有统计学意义(P<0.05).与空白组相比,模型组caspase-8、caspase-3阳性表达率均升高,差异均有统计学意义(P<0.05);与模型组比较,桂药生精胶囊组、阳性对照组caspase-8、caspase-3阳性表达率均降低,差异均有统计学意义(P<0.05);与阳性对照组比较,桂药生精胶囊组caspase-3阳性表达率降低,差异有统计学意义(P<0.05),caspase-8阳性表达率差异无统计学意义(P>0.05).结论 桂药生精胶囊可通过下调caspase-8、caspase-3蛋白表达,抑制生精细胞凋亡,进而改善少弱精子症模型大鼠睾丸组织病理损伤,改善生精细胞分化水平,促进大鼠生殖功能恢复.
Abstract
Objective To observe the effect of Guiyao Shengjing Capsules on the expression of caspase-8 and caspase-3 in model rats of oligoasthenozoospermia.Methods Forty male SD rats were divided into blank group(n=10)and model group(n=30)according to the random number table method.The model group was constructed by intraperitoneal injection of cyclophosphamide to construct oligoasthenospermia model.After the model was successfully prepared,the model group was divided into model group,Guiyao Shengjing capsules group and positive control group according to the random number table method,with 10 rats in each group.The positive control group was given L-carnitine oral so-lution 200 mg/(kg·d)by gavage,and the Guiyao Shengjing capsules group was given Guiyao Shengjing capsules 52.5 mg/(kg·d),the blank group and the model group were given the same amount of 0.9%sodium chloride solution by gavage,once a day,for 28 days.24 h after the last administration,testis and epididymis were extracted,sperm quality was detected,testicular tissue morphology was observed under light micro-scope,and mRNA and protein expressions of caspase-8 and caspase-3 in testicular tissue were detected by quantitative reverse transcription pol-ymerase chain reaction and immunohistochemistry.Results Compared with the blank group,the number and activity of sperm in the model group decreased,and the difference was statistically significant(P<0.05).Compared with the model group,the sperm number and motility of the rats in the Guiyao Shengjing Capsules group and the positive control group increased,and the differences were statistically significant(P<0.05).Compared with the blank group,the testicular seminiferous tubules of the rats in the model group were atrophied,the degree of differentiation of spermatogenic cells was significantly reduced,the number of sperm was significantly reduced,and vacuolization was observed.Compared with the model group,the morphological structure of testicular seminiferous tubules in the Guiyao Shengjing Capsules group and the positive control group was improved,the degree of differentiation of spermatogenic cells was increased,the number of sperm was significantly increased,and the outer membrane was repaired.Compared with the blank group,the expression levels of caspase-8 and caspase-3 mRNA in the model group were in-creased,and the differences were statistically significant(P<0.05).Compared with the model group,the expression levels of caspase-8 and caspase-3 mRNA in the Guiyao Shengjing Capsules group and the positive control group were decreased,and the differences were statistically sig-nificant(P<0.05).Compared with the blank group,the positive expression rates of caspase-8 and caspase-3 in the model group were in-creased,and the differences were statistically significant(P<0.05).Compared with the model group,the positive expression rates of caspase-8 and caspase-3 in the Guiyao Shengjing Capsules group and the positive control group were decreased,and the differences were statistically sig-nificant(P<0.05).Compared with the positive control group,the positive expression rate of caspase-3 in the Guiyao Shengjing Capsules group decreased,and the difference was statistically significant(P<0.05);there was no statistically significant difference in the positive expression rate of caspase-8(P>0.05).Conclusion Guiyao Shengjing Capsules can reduce the apoptosis of spermatogenic cells by down-regulating the expression of caspase-8 and caspase-3 proteins,thus improving the pathological damage of testicular tissue in oligozoospermia model rats,improving the differentiation level of spermatogenic cells and promoting the recovery of reproductive function in rats.
