Comparison on the Purification of Gleditsia sinensis Saponin by the H2O2 Decolorization and Column Chromatography
Commercially available G.sinensis saponin(GSS)had deep color,heavy odor,and low saponin content,which limited its application.In this paper,the solvent extraction of GSS was separated and purified by H2O2 decolorization and AB-8 macroporous adsorption resin column chromatography.The purity,structure composition,surface activity,foam and emulsification properties of saponin were compared and studied.The results showed that the structure of saponin purified by H2O2 decolorization(H2O2-GSS)was damaged,the saponin content was 48.02%,but the foam stability was excellent and the half-life(t1/2)was 10 h.Results of column chromatography showed that the highest purity of saponin was achieved through elution using 60%ethanol,resulting in a 92.12%saponin mass fraction.Additionally,the foam performance was the best,reaching a height of up to 11 cm,and the emulsion was the most stable within 24 h.After 24 hours,stratification occurred at a height of 2.2 cm.The mass fractionofsaponinelutedby75%ethanolwas86.40%,thesurfacetensionvaluewasthelowest(41.50 mN/m),and the critical micelle concentration(CCMC)was 3 g/L(having the best surface activity).The H2 O2-GSS showed better foam stabilization ability.The GSS obtained by column chromatography had higher purity,better surface activity,foam performance and emulsion stability.The surface activity of purified recombinant saponin(M-GSS)was further improved,which provided ideas for the development of high-performance surfactants.
