Antioxidant,Hypoglycemic and Hypolipidemic Activities in vitro of Different Polar Solvent Extracts from Dendrocalamus giganteus Leaves
The 70%ethanol extract from Dedrocalamus giganteus leaves was fractionated by successive extraction with petroleum ether,dichloromethane,ethyl acetate and n-butanol by graded extraction to obtain five different polar solvent extracts,including petroleum ether extract(PEE),dichloromethane extract(DME),ethyl acetate extract(EAE),n-butanol extract(BE),and water extract(WE),respectively.The contents of total flavonoids and polyphenols were determined,and the antioxidant,hypoglycemic and hypolipidemic activities of the five extracts in vitro were also evaluated.The results showed that the total flavonoid and polyphenols mass fractions of the ethyl acetate extract were the highest with(195.14±0.46)and(52.01±0.39)mg/g,respectively which were significantly higher than those of the other extracts(p<0.05).Furthermore,all the different polar solvent extracts showed a certain antioxidant activity,hypoglycemic and hypolipidemic abilities in vitro.Among them,the ethyl acetate extract had the strongest scavenging activities on ABTS+·,inhibiting activities on α-glucosidase and α-amylase,and the half inhibitory concentration(ICs0)values were(118.73±0.29),(45.74±3.31)and(53.81±2.59)mg/L,respectively.Meanwhile,the binding rates of the ethyl acetate extract with sodium glycinate and sodium taurocholate were the highest at the same concentration,which reached(64.06±0.18)%and(59.27±0.35)%,at a mass concentration of 0.25 g/L,respectively.Polyphenol content of different extracts of D.giganteus leaves showed significant positive correlation with DPPH·and ABTS+·scavenging abilities(p<0.05),with correlation coefficients of 0.94 and 0.81,respectively.The content of total flavonoids showed significant positive correlation with hypoglycemic and hypolipidemic ability in vitro(p<0.05),with correlation coefficients were 0.75-0.84.
