Chemical Composition Analysis of Extracts From Phoebe zhennan Wood Obtained by Different Extraction Methods
Taking Phoebe zhennan S.K.Lee & F.N.Wei as the research object, the P.zhennan extracts were obtained by the five methods of steam distillation(SD)method, supercritical CO2 extraction(SFE)method, ethanol extraction(EE)method, ethyl acetate extraction(EAE)method and n-hexane extraction(HE)method. The differences of chemical composition of P.zhennan were compared by gas chromatography and mass spectrometry(GC-MS),and the main chemical components were quantitatively determined by high performance liquid chromatography(HPLC).The results showed that a total of 132 compounds were identified in the extracts obtained by the five methods, and the sum of their relative peak areas ranged from 88.49%to 97.16%.The chemical components mainly included esters, alcohols, hydrocarbons, alkanes, acids, aldehydes, ketones and phenols.Among the P.zhennan extracts obtained by different extraction methods, there were three kinds of common compounds:alcohols, alkenes and ketones, where alcohols were the important part of all extracts.Agarospirol and valerianol were decisive compounds in all P.zhennan extracts. The sum of the relative peak areas of P.zhennan extracts by the five methods ranged from 13.73%to 29.43%,and there was a very significant positive correlation between them.Due to their similar chemical structures and the same polar groups, the retention behavior of the two compounds in the chromatography was basically the same, and the separation was difficult.By supercritical CO2 extraction method, the yields of agarospirol and valerianol were the highest,0.305%and 0.157%,respectively,followed by steam distillation method, and solvent extraction method was the lowest.
