目的:采用动态功能连接分析技术探讨孤独症谱系障碍(ASD)患者右侧梭状回与全脑动态功能连接特点,并分析异常脑区的动态功能连接时间变异性与ASD主要症状的相关性.方法:将孤独症脑影像公开数据库ABIDE I和Ⅱ中美国纽约大学朗格尼医学中心收集的120 例ASD患者和131 名与之年龄相匹配的正常对照作为研究对象,利用滑动窗法,计算右侧梭状回与全脑动态功能连接的时间变异性,采用双样本t检验,以年龄、性别、利手及智商作为协变量,比较两组被试右侧梭状回与全脑动态功能连接时间变异性的差异,并将差异脑区动态功能连接时间变异值与ASD主要症状评分进行Spearman相关分析.结果:与正常对照相比,ASD组的右侧梭状回与右侧额中回、双侧内侧额中回、左侧背外侧额上回的动态功能连接时间变异性显著升高(Gaussian Random Field,GRF校正,体素水平P<0.005 且团块水平P<0.05),差异脑区的动态功能连接时间变异值与ASD诊断观察量表(ADOS)重复刻板行为分数呈正相关(P<0.05).结论:ASD患者右侧梭状回与全脑动态功能连接变异值增高的脑区均属于社会脑组成部分,该发现为社会脑功能异常在ASD形成机制的研究中提供了新的依据.
Temporal variability of dynamic brain functional connectivity in the resting state of the fusiform gyrus in autism spectrum disorders
Objective:To explore the characteristics of dynamic functional connectivity between the right fusiform gyrus and the whole brain in patients with autism spectrum disorder(ASD)using dynamic functional connectivity analysis technology,and to analyze the correlation between the temporal variability of dynamic func-tional connectivity in abnormal brain regions and the main symptoms of ASD.Method:A total of 120 ASD patients and 131 age-matched normal controls collected from the Langley Medical Center of New York University in the publicly available databases ABIDE I and II of autism brain imaging were used as the research subjects.The sliding window method was used to calculate the temporal variability of the dynamic functional connectivity between the right fusiform gyrus and the whole brain.Two sample t-tests were used,with age,gender,dexterity,and intelligence as covariates Results:Compare the differences in temporal variability of dynamic functional connectivity between the right fusiform gyrus and the whole brain between two groups of participants,and per-form Spearman correlation analysis between the temporal variability of dynamic functional connectivity in differ-ent brain regions and the main symptom score of ASD.Compared with the normal control group,the variability of dynamic functional connectivity time between the right fusiform gyrus and the right middle frontal gyrus,bilateral medial middle frontal gyrus,and left dorsolateral superior frontal gyrus in the ASD group was significantly in-creased(Gaussian Random Field,GRF corrected,voxel level P<0.005 and mass level P<0.05),The varia-tion of dynamic functional connectivity time in different brain regions was significantly correlated with the repeat-ed stereotyped behavior score on the Autism Diagnostic Observation Schedule(ADOS)(P<0.05).Conclusion:The brain regions with increased variability in the right fusiform gyrus and global dynamic function-al connectivity in ASD patients belong to the social brain component.This finding provides new evidence for the study of social brain dysfunction in the formation mechanism of ASD.
