Objective To investigate the impact of LINC01140 overexpression on the proliferation and invasion of lung adenocarcinoma cell H1975,so as to provide a basis for the selection of targeted drugs and prognostic evaluation. Methods Based on the results of bioinformatics, lung adenocarcinoma cell line H1975 with positive mutation of epidermal growth factor receptor(EGFR)T790M was selected and overexpressed LINC01140 cell line was constructed by transfection technique, and divided into a control group(H1975/NC)and a high expression group(H1975/LINC01140).The expression level of LINC01140 in the cell line was detected by RT-PCR.They were divided into control group(H1975/NC)and high ex-pression group(H1975/LINC01140).Cells in the two groups were cultured. The proliferation ability of cells was detected by MTS test, the migration ability of cells was detected by wound healing assay, and the invasion ability of cells was detected by Transwell test. Results The results of bioinformatics revealed that the expression of LINC01140 in lung cancer tissues was lower than that in adjacent tissues, which was related to clinical features and survival(P<0.05).In the meantime, the re-sults of this study confirmed the successful construction of LINC01140 overexpressed H1975 cell line.Compared with H1975/NC,the expression level of LINC01140 in H1975/LINC01140 increased, cell absorbance and migration rate increased, and the number of transmembrane cells increased(P<0.05).Conclusion LINC01140 overexpression promotes the prolifera-tion,migration and invasion of lung adenocarcinoma cell H1975.Detection of its expression level can guide the selection of clinical targeted drugs, improve efficacy, and thus improve prognosis.
