Influence of temperature and time on Lewis blood group antibody and analysis of gene polymorphism
Objective:To investigate the effects of different reaction temperature and time on Lewis blood group antibody and the characteristics of gene polymorphism.Methods:A total of 35 blood donors with Lewis an-tibody identified serologically from January 2020 to December 2022 were selected,was used to carry out the exper-iments on the influence of different temperature and time,and the Leiws antigen was identified.According to the results of Lewis antigen serotyping,the FUT2 and FUT3 genes were detected by PCR-SBT technology to confirm the experimental results.Results:The 35 cases of Lewis antibodies were agglutinated with different intensities in the saline test tube method,and the antibody specificity was identified as 18 cases of anti-Leb,6 cases of anti-Lea,and 11 cases of anti-Lea combined with anti-Leb.The experimental results showed that the optimal condition for Lewis antibody detection was 22℃ for 15 min.The serological phenotypes of Lewis antigen were Le(a+b-)in 8 cases,Le(a-b+)in 6 cases,and Le(a-b-)in 21 cases.Conclusion:By exploring the effects of temperature and time on Lewis blood group system antibodies,the detection rate can be improved and the risk of weak antibod-ies being missed can be reduced.Serology combined with molecular biology technology was used to identify the Lewis antigen phenotype,and it was found that the molecular inheritance of the blood group system had abundant gene polymorphisms in Changzhou,and 3 cases of combined mutations were not included,so as to establish a gene polymorphism database of Lewis blood group system in the blood donor population in this region and provide a ba-sis for clinical precision blood transfusion.
