基于IL-6/STAT3通路研究苗药方剂"四大血"对胶原诱导性关节炎小鼠的保护作用

目的 基于白细胞介素-6(IL-6)/STAT3 通路研究苗药方剂"四大血"(SX)对胶原诱导性关节炎(CIA)小鼠的保护作用机制。方法 取40只C57BL/6雄性小鼠随机均分为4 组[正常组、模型组、SX组和甲氨蝶呤(MTX)组]，采用鸡Ⅱ型胶原和完全弗氏佐剂混合乳化剂诱导类风湿性关节炎(RA)小鼠模型。造模成功后按照实验分组(每组 6 只)进行灌胃给药。分别检测各组小鼠的足肿胀程度、关节病理改变、关节炎指数(AI)评分、肿瘤坏死因子-α(TNF-α)和IL-6 水平、关节滑膜组织中p-STAT3 蛋白表达情况。结果 SX对RA疾病相关指标有显著改善作用，可以降低小鼠足肿胀程度、关节炎滑膜炎症、AI评分、TNF-α和IL-6 水平，抑制p-STAT3 蛋白的表达。结论 SX可以发挥明显的抗RA作用，其可通过抑制IL-6/STAT3 信号通路蛋白(p-STAT3)表达发挥对CIA小鼠的保护作用。
Study on the protective effect of the Miao ethnomedicine recipe"Sidaxue"on collagen induced arthritis mice based on IL-6/STAT3 pathway
Objective To study the protective effect and mechanism of the Miao ethnomedicine recipe"Sidaxue"(SX)on collagen induced arthritis(CIA)mice based on the interleukin-6(IL-6)/STAT3 pathway.Methods Forty male C57BL/6 mice were randomly divided into 4 groups[normal group,model group,SX group and methotrexate(MTX)group],and the rheumatoid arthritis(RA)mouse model was induced by combination of chicken typeⅡ collagen and complete Freund's adjuvant.After successful modeling,the drug was administered by gavage according to experimental groups(6 mice in each group).The degree of foot swelling,joint pathological changes,Arthritis Index(AI)score,tumor necrosis factor-α(TNF-α)and IL-6 levels,and p-STAT3 protein expression in joint synovium were detected respectively.Results SX can significantly improve RA disease-related indicators,reduce the degree of foot swelling,arthritis synovitis,AI score,TNF-α and IL-6 levels,and inhibit p-STAT3 protein expression.Conclusion SX can exert a significant anti-RA effect and exert a protective effect on mice with CIA by inhibiting the expression of IL-6/STAT3 signaling pathway protein(p-STAT3).
