Effects of vitamin K2 on bone tissue and glucose metabolism in diabetic osteoporosis rats
Objective To investigate the effects of vitamin K2 on bone tissue and glucose metabolism in diabetic osteoporosis(DOP)rats.Methods Eighteen 12-week-old male SD rats were randomly divided into normal group,model group and intervention group.The normal group was not modeled,and the DOP model was constructed in the model group and the intervention group.After successful modeling,the intervention group was given vitamin K2 by gavage,and the model group and the normal group were given the same amount of normal saline by gavage.After 4 weeks of intervention,the femurs of rats were separated and collected,and the bone mineral density(BMD)and bone morphology related parameters of distal femur were detected by Micro-CT;the number of osteoclasts in rats was observed by immunohistochemical staining;the number of osteoblasts in rats was observed by immunofluorescence staining;the mechanical properties of bone tissue in rats were observed by mechanical detector;the levels of fasting insulin(FINS)and fasting plasma glucose(FPG)were measured in tail venous blood of rats;homeostasis model assessment of insulin resistance(HOMA-IR)and insulin sensitive index(ISI)were measured by homeostasis model assessment.Results The BMD,bone volume fraction,trabecular number and trabecular thickness of the intervention group were greater than those of the model group(P<0.05).The results of immunohistochemistry and immunofluorescence staining showed that the number of osteoclasts decreased significantly after vitamin K2 supplementation,and the number of osteoblasts increased significantly compared with the model group.The ultimate load and stiffness of the intervention group were higher than those of the model group(P<0.05).The FINS,FPG levels and HOMA-IR in the intervention group were lower than those in the model group,ISI was higher than that in the model group(P<0.05).Conclusion Vitamin K2 can significantly slow down the bone loss of DOP rats and improve the glucose metabolism indexes,which provides a new idea for the treatment of DOP.
