Discussion the mechanism of moist exposed burn ointment in the treatment of diabetic foot based on bioinformation technology
Objective To explore the mechanism of moist exposed burn ointment(MEBO)in the treatment of diabetic foot(DF)through network pharmacology.Methods The active chemical constituents and active ingredient target information of MEBO were obtained through Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP),and the target gene name was obtained through UniProt database;the target genes related to DF were queried through Human Gene Comprehensive Database(GeneCards),and the target of MEBO active ingredient was mapped with the target of DF by Venny2.1 to obtain the potential target of action.The intersection targets were imported into the STRING website for analysis,and the protein-protein interaction(PPI)network map was constructed using Cytoscape3.10.0 to select key target genes.The intersection targets were imported into the GO and KEGG pathway enrichment analysis.The"drug-ingredient-target"network mdp was constructed using Cytoscape software.Results The main chemical components of MEBO in the treatment of DF were berberine,wogonin,juteoside A,coptisine,obacunone,etc.The key role targets were epidermal growth factor receptor(EGFR),nitric oxide synthase 3(NOS3),protein kinase B1(AKT1),interleukin-1B(IL-1B),interleukin-6(IL-6),tumor necrosis factor(TNF),etc.The key role pathways were AGE-RAGE,HIF-1,VEGF,NF-κB and other signaling pathways.Conclusion This study initially revealed that the active ingredients of MEBO can treat DF from multiple targets and multiple pathways,play a role in improving the microenvironment,inhibiting wound infection and promoting tissue repair and regeneration.
