Objective To explore the effect of dietary intervention on the susceptibility of offspring diabetes in gestational diabetes mellitus(GDM)mice. Methods Seventy-two 3-week-old healthy female mice were selected and randomly divided into GDM group and low-fat(LF)group.The GDM group was fed with high-fat(HF)diet to establish a GDM mouse model, and the LF group was fed with LF diet. The offspring of the two groups of mice were fed with HF diet and LF diet after weaning at 3 weeks of age,and were divided into GDM HF(G-HF)group,GDM LF(G-LF)group,LF HF(L-HF)group and LF LF(L-LF)group.After intervention, the body weight and fasting blood glucose(FBG)were measured, and insulin tolerance test(ITT)was conducted. Results The success rate of GDM mouse model induced by HF diet was 33.33%,the pathogenesis of this model was similar to the pathophysiological development of human GDM.At 6 weeks of dietary intervention, the body weight of the G-HF group was higher than that of the other three groups, and the body weight of the L-HF group was higher than that of the L-LF group(P<0.05);after 12 weeks of diet intervention, the body weight of the G-HF group was higher than that of the other three groups(P<0.05).After 6 weeks of diet intervention, the FBG level of the G-HF group was higher than that of the G-LF group and the L-LF group, and the FBG level of the L-HF group was higher than that of the L-LF group(P<0.05);at 12 weeks of diet intervention, the FBG level of the G-HF group was higher than that of the other three groups, and the FBG level of the L-HF group was higher than that of the L-LF group(P<0.05).At 7 weeks of age,the sensitivity of the L-LF group to insulin was higher than that of the G-HF group. Conclusion GDM offspring have obvious glucose metabolism disorder and weight gain, and LF diet can reduce the risk of diabetes in GDM offspring.
