Preliminary screening research of differentially expressed proteins between low grade glioma and normal brain tissue based on iTRAQ technology
Objective To screen differentially expressed proteins between low grade glioma and normal brain tissue by isobaric tags for relative and absolute quantization(iTRAQ)technology.Methods The total protein was extracted from mixing 6 low grade glioma solid tissue samples and 2 normal brain tissue samples for quantitative analysis and enzymatic hydrolysis.Liquid chromatography coupled with tandem mass spectrometry(LC-MS/MS)analysis was performed after iTRAQ labeling.The correlation spectra were analyzed by Mascot2.9 software.Results A total of 162 differentially expressed proteins were identified between low grade glioma and normal brain tissue,including 34 up-regulated proteins and 128 down-regulated proteins.These differentially expressed proteins had a variety of biological activities and participated in different metabolic and signaling pathways.Conclusion iTRAQ technology can effectively identify many differentially expressed proteins between low grade glioma and normal brain tissue,so as to lay a foundation for the subsequent search for biomarkers of low grade glioma.
