基于系统药理学探索中药生脉饮有效成分的药理作用机制

目的 基于系统药理学方法探究中药生脉饮活性成分、靶点及疾病之间的复杂关系。方法 在中药系统药理学数据库分析平台(TCMSP)数据库,以口服生物利用度(OB)(人参和五味子OB≥60%,麦冬OB≥45%)和类药性(DL)≥0。1 筛选生脉饮的活性成分、预测作用靶点,构建活性成分-靶点-疾病网络。结果 筛选出活性成分 17 个,其中人参 11 个成分,麦冬 2 个成分,五味子 4 个成分。共获得靶点 79 个,靶点作用的疾病13种。网络图中活性成分和靶点共有176 条边相连,即平均每个活性成分对应约10。3 个靶点,平均每个靶点与4。6个活性成分相关联。靶点和疾病共235 条边相连。结论 本研究直观地构建了生脉饮活性成分、靶点蛋白和疾病的相互作用,从而揭示生脉饮在多种临床疾病中的功效,为进一步研究生脉饮作用机制提供理论依据。
Explore the pharmacological mechanism of effective components of Shengmaiyin based on system pharmacology
Objective To explore the complex relationship of the active ingredients,targets and disease of Shengmaiyin based on systematic pharmacology method.Methods The active ingredients of Shengmaiyin were screened and the targets were predicted on traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP)with oral bioavailability(OB)(Renshen and Wuweizi OB≥60%,Maidong OB≥45%)and drug-likeness(DL)≥0.1,and the active ingredient-target-disease network was constructed.Results A total of 17 active ingredients were screened,including 11 components of Renshen,2 components of Maidong,and 4 components of Wuweizi.A total of 79 targets and 13 diseases were obtained.The active ingredients and targets in the network diagram were connected by 176 edges,that is,each active ingredient corresponded to about 10.3 targets on average,and each target was associated with 4.6 active ingredients on average.The target and the disease are connected by a total of 235 edges.Conclusion This study intuitively constructed the interaction between the active ingredients,target proteins and diseases of Shengmaiyin,thus revealing the efficacy of Shengmaiyin in a variety of clinical diseases,and providing theoretical basis for further research on the mechanism of Shengmaiyin.
