基于"血不利则为水"理论探讨当归芍药散调控妊娠大鼠凝血功能及改善血栓前状态的机制研究

目的 基于"血不利则为水"理论探讨当归芍药散调控妊娠大鼠凝血功能及改善血栓前状态的作用机制。方法 选取 60 只 8 周龄SD雌性大鼠,随机将其分为空白组、模型组、中药低剂量组[0。07 g/(100 g·d)]、中药中剂量组[0。14 g/(100 g·d)]、中药高剂量组[0。28 g/(100 g·d)]、西药组[20 IU/(100 g·d)],每组 10 只,均与健康雄性大鼠交配,妊娠第 1 天用盐酸肾上腺素加冰水法建立血栓前状态妊娠大鼠模型,造模过程中模型组2只大鼠死亡。比较六组的存活胚胎数、丢失胚胎数、胚胎丢失率;比较六组的纤维蛋白原(FIB)、抗凝血酶-Ⅲ(AT-Ⅲ)、D-二聚体(DD)和纤溶酶原(PLG)水平。结果 模型组的存活胚胎数低于空白组,丢失胚胎数和胚胎丢失率高于空白组,差异具有统计学意义(P<0。05);中药中剂量组、中药高剂量组、西药组的存活胚胎数高于模型组,差异具有统计学意义(P<0。05);中药高剂量组、西药组的丢失胚胎数低于模型组,差异具有统计学意义(P<0。05);中药中剂量组、中药高剂量组、西药组的胚胎丢失率低于模型组,差异具有统计学意义(P<0。05)。模型组的FIB、DD和PLG水平高于空白组,差异具有统计学意义(P<0。05);中药低剂量组、中药中剂量组、中药高剂量组、西药组的FIB、DD和PLG水平低于模型组,AT-Ⅲ水平高于模型组,差异具有统计学意义(P<0。05)。结论 当归芍药散能够降低血栓前状态模型大鼠的丢失胚胎数、胚胎丢失率及DD、PLG、FIB水平,提高存活胚胎数及AT-Ⅲ水平,保障胚胎的正常发育,改善血液高凝状态,促进微循环,从而达到抗血栓作用。
Explore the mechanism of Danggui Shaoyao powder in regulating coagulation function and improving prethrombotic state in pregnant rats based on the theory of"unfavorable blood is water"
Objective To explore the mechanism of Danggui Shaoyao powder in regulating coagulation function and improving prethrombotic state in pregnant rats based on the theory of"unfavorable blood is water".Methods A total of 60 8-week-old SD female rats were selected and randomly divided into blank group,model group,low-dose Chinese medicine group[0.07 g/(100 g·d)],medium-dose Chinese medicine group[0.14 g/(100 g·d)],high-dose Chinese medicine group[0.28 g/(100 g·d)]and western medicine group[20 IU/(100 g·d)],with 10 rats in each group,all rats were mated with healthy male rats.On the first day of pregnancy,the prethrombotic state pregnancy rat model was established by adrenaline hydrochloride and ice water method,during the modeling process,2 rats in the model group died.The number of surviving embryos,the number of lost embryos and the rate of embryo loss were compared among the six groups;the levels of fibrinogen(FIB),antithrombin-Ⅲ(AT-Ⅲ),D-dimer(DD)and plasminogen(PLG)were compared among the six groups.Results The number of surviving embryos in the model group was lower than that in the blank group,and the number of lost embryos and the rate of embryo loss were higher than those in the blank group,and the differences were statistically significant(P<0.05);the number of surviving embryos in the medium-dose Chinese medicine group,high-dose Chinese medicine group and western medicine group was higher than that in the model group,and the difference was statistically significant(P<0.05);the number of lost embryos in the high-dose Chinese medicine group and western medicine group was lower than that in the model group,and the difference was statistically significant(P<0.05);the rate of embryo loss of the medium-dose Chinese medicine group,high-dose Chinese medicine group and western medicine group was lower than that of the model group,and the difference was statistically significant(P<0.05).The levels of FIB,DD and PLG in the model group were higher than those in the blank group,and the differences were statistically significant(P<0.05);the levels of FIB,DD and PLG in the low-dose Chinese medicine group,medium-dose Chinese medicine group,high-dose Chinese medicine group and western medicine group were lower than those in the model group,and the level of AT-Ⅲ was higher than that in the model group,and the differences were statistically significant(P<0.05).Conclusion Danggui Shaoyao powder can reduce the number of lost embryos,the rate of embryo loss and the levels of DD,PLG,FIB in prethrombotic state model rats,increase the number of surviving embryos and AT-Ⅲ level,ensure the normal development of embryos,improve blood hypercoagulability,promote microcirculation,and achieve antithrombotic effect.
