Role of cold shock protein RBM3 in renal ischemia reperfusion injury in young rats
Objective To establish a model of renal ischemia reperfusion injury(RIRI)in young rats,and to explore the effect of cold shock protein RBM3 induced by mild hypothermia on RIRI.Methods Twenty-four 7 d SD young rats were randomly divided into normal control group(NC group),sham operation group(Sham group),RIRI non-hypothermia group(NH group)and RIRI hypothermia group(TH group),with 6 rats in each group.Renal RIRI model was established and whole-body mild hypothermia therpy was performed;the levels of serum creatinine(SCr)and Cystatin C(CysC)were detected;the left renal tissue was taken for HE staining to observe the pathological changes,and the right renal tissue was taken to detect the expression of YAP1 and NRF2 genes by qPCR;the expression of RBM3 protein was detected by Western Blot.Results There were no significant differences in the levels of SCr and CysC between the NC group and the Sham group(P>0.05);the levels of SCr and CysC in the NH group and the TH group were significantly higher than those in the NC group,and the levels of SCr and CysC in the NH group were significantly higher than those in the TH group(P<0.05).The pathological tissue score of the NH group and the TH group was higher than that of the NC group,and the pathological tissue score of the NH group was higher than that of the TH group(P<0.05).The expression of YAP1 and NRF2 genes in the TH group were significantly higher than those in the NC group and the NH group(P<0.05).The expression of RBM3 protein in the TH group was significantly higher than that in the NC group and the NH group(P<0.05).Conclusion RBM3 induced by systemic mild hypothermia has a protective effect on RIRI in young rats.This protective effect may be related to the reduction of inflammatory response by RBM3 induced by hypothermia.
