Expression and clinical significance of miR-6736-3p in gastric adenocarcinoma tissues
Objective To investigate the expression and clinical significance of miR-6736-3p in gastric adenocarcinoma tissues. Methods The gastric adenocarcinoma tissues and adjacent normal tissues of 73 patients were collected, the expression of miR-6736-3p in gastric adenocarcinoma tissues and adjacent normal tissues was detected by qRT-PCR,and the relationship between the expression level of miR-6736-3p and the clinicopathological features and prognosis of gastric adenocarcinoma was analyzed. The growth and proliferation of gastric adenocarcinoma were detected by plate cell clone formation assay and CCK-8 assay. Results The results of qRT-PCR showed that the expression level of miR-6736-3p in the gastric adenocarcinoma tissues was higher than that in the adjacent normal tissues(P<0.05);the expression level of miR-6736-3p in the gastric adenocarcinoma AGS cells was higher than that in the GES-1 cells(P<0.05).The expression level of miR-6736-3p was closely related to tumor size, differentiation degree and prognosis(P<0.05).In vitro cell experiments showed that the number of cell clone formation and optical density(OD)value at 72 h in the miR-6736-3p overexpression group were higher than those in the control group(P<0.05);the number of cell clone formation and OD value at 72 h in the miR-6736-3p decreased expression group were lower than those in the control group(P<0.05).Conclusion miR-6736-3p is highly expressed in gastric adenocarcinoma tissues, which is closely related to tumor size and differentiation degree. It may be an oncogenic factor that can promote the growth and proliferation of gastric adenocarcinoma.
