Study on relationship between the expression of serum ZEB1 and USP22 and glucose lipid metabolism and insu-lin resistance in patients with type 2 diabetes
Objective To detect the expression levels of serum zinc finger E-box-binding protein 1(ZEB1)and ubiquitin-specific protease 22(USP22)genes in patients with type 2 diabetes,and analyze the relationship between them and glucose and lipid metabolism and insulin resistance.Methods From June 2020 to June 2023,120 patients with type 2 diabetes diagnosed and treated in the First Affiliated Hospital of Suzhou University were collected as the study subjects(diabetes group).meantime,120 healthy people who underwent physical examination were as the control group.The qRT-PCR method was applied to detect the expression levels of ZEB1 mRNA and USP22 mRNA in serum;Pearson method was applied to analyze the correlation between the expression levels of ZEB1 mRNA and USP22 mRNA in serum of type 2 diabetes patients,and the correlation between them and BMI,TG,TC,FPG,FIns,HOMA-IR,ISI,HO-MA-β;multiple linear regression was applied to analyze the factors influencing the expression of ZEB1 mRNA and USP22 mRNA in serum of type 2 diabetes patients.Results The levels of BMI,TG,TC,FPG,FIns,HOMA-IR,ZEB1 mRNA and USP22 mRNA in the diabetes group were obviously higher than those in the control group,while ISI and HO-MA-β were obviously lower than those in the control group(P<0.05).Pearson correlation analysis showed that there was a positive correlation between the expression of ZEB1 mRNA and USP22 mRNA in serum of type 2 diabetes patients(r=0.425,P<0.001);the expression levels of serum ZEB1 mRNA and USP22 mRNA were positively correlated with FPG,FIns,HOMA-IR(P<0.05),and negatively correlated with ISI and HOMA-β(P<0.05).Multiple linear regres-sion analysis showed that an increase in FIns and a decrease in ISI were the influencing factors of serum ZEB1 mRNA expression level(P<0.05);an increase in FPG was a factor influencing the expression level of USP22 mRNA(P<0.05).Conclusions There is a positive correlation between the expression of ZEB1 mRAN and USP22 mRAN in the serum of type 2 diabetes patients,and the both are correlated with some indicators of glucose and lipid metabolism and insulin resistance.
