Optical Conditions for Solid-state Fermentation of the Biocontrol Fungal AH-1 Based on Response Surface Methodology
[Objective]In order to develop new biocontrol agent,the conditions of solid-state fermentation of biocontrol strain AH-1 were explored and optimized.[Methods]Singal factor assessment and response surface methodology were performed.[Result]The results showed that the three main factors affecting solid-state fermentation were inoculation amount,wheat bran content and temperature.The optimized solid-state fermentation medium composition was wheat bran:corn flour of 3:2.The optimized solid-state fermentation culture conditions were as follows:initial pH-value of 7,water content of 60%,spore suspension inoculation amount of about 18%(with concentration of 1×106 conidia/mL),temperature of 30℃,and fermentation period of 10 d.Under these conditions,the conidial yield was 1.672×1010 conidia/g,which was about 2~3 times higher than that of the pre-optimization factor test.[Conclusion]The fermentation conditions optimized by the response surface method could obviously increase the sporulation of biocontrol strain AH-1 in solid-state fermentation,which laid a foundation for its large-scale production and preparation of microbial agents.
