Objective:To screen differentially expressed long non-coding RNA and conduct a bioinformatics analy-sis in 2 h mild hypothermia mediated neuroprotection after resuscitation from Wistar rats cardiac arrest.Methods:Cere-bral cortex from ventricular-fibrillation-model male Wistar rats(n=10)randomized into equal MH and control normother-mia groups at 2 h post-resuscitation was analyzed for expression.LncRNA expression evaluated via Agilent lncRNA mi-croarray was validated by real-time reverse-transcription polymerase chain reaction.Results:In MH,6 lncRNAs were upregulated(>2-fold)and 21 were downregulated(<0.5-fold),9 mRNAs were upregulated(>2-fold)and 11 were downregulated(<0.5-fold)at 2 h post-resuscitation compared with CN.Significantly changed lncRNAs co-expressed with mRNAs involving hundreds of GO and KEGG annotated clusters,predominantly involving neuronal apoptosis,oxida-tive stress,metabolism,and protein synthesis,transport,and degradation pathways.Conclusion:MH significantly al-ters cerebral lncRNA expression profiles,potentially co-regulating neuronal apoptosis-associated mRNA to effect hypo-thermia-mediated neuroprotection.
