摘要

目的:采用两样本孟德尔随机化(Mendelian Randomization,MR)方法,探讨高尿酸与房颤之间的因果关系.方法:从IEU Open GWAS数据库中检索单核苷酸多态性(SNP)数量最多的文献数据,将高尿酸作为暴露因素,将房颤作为结局变量.采用MR-Egger法、逆方差加权法(IVW)进行孟德尔随机化分析,研究高尿酸与房颤的因果关系.结果:去除连锁不平衡影响后,最终确定了3个独立且高度相关的SNPs,的r²值<0.01,与高尿酸水平的关联显著(P值小<5×108).IVW法估计高尿酸与房颤的因果效应,显示两者存在正向的因果联系(OR=1.745,95%CI为1.012-1.355,P=0.033).结论:高尿酸与房颤之间存在显著的因果关系,对潜在房颤的高尿酸患者实行早期诊断和干预是必要的.

Abstract

Objective:Two sample MR was used to investigate the causal relationship between high uric acid and atrial fibrillation.Methods:Literature data with the largest number of SNPS were retrieved from IEU Open GWAS data-base.High uric acid was used as exposure factor and atrial fibrillation was used as outcome variable.The MR-Egger method and IVW method were used to conduct MR to study the causal relationship between high uric acid and atrial fi-brillation.Results:After removing the effects of linkage disequilibrium,three independent and highly correlated SNPs were finally identified with an r² value<0.01,which was significantly associated with high uric acid level(P value<5×108).IVW method showing a positive causal association(OR=1.745,95%CI 1.012-1.355,P=0.033).Conclusion:There is a significant causal relationship between high uric acid and atrial fibrillation,and it is necessary to implement early diagnosis and intervention for high uric acid patients with potential atrial fibrillation.
