摘要

目的 初步研究三黄颗粒退黄作用的机制.方法 应用α-萘异硫氰酸酯(ANIT)致小鼠胆汁瘀积模型,将小鼠随机分为6组:空白组,ANIT模型组,茵栀黄组,三黄颗粒低、中、高剂量组,各组小鼠均灌胃(ig)给药10d.给药第8天除正常对照组外,其余各组小鼠均ig ANIT 70 mg/kg,之后继续ig给药,48 h后断头取血,检测其血清中的丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)、总胆红素(STB)、碱性磷酸酶(ALP),并剖腹取肝脏,称取肝脏重量,计算其肝指数;用四氯化碳( CC14)致小鼠肝细胞损伤模型,将小鼠随机分为7组:空白组,ANIT模型组,联苯双酯组,茵栀黄组,三黄颗粒低、中、高剂量组,各组小鼠均ig给药10 d,末次给药后2h,除正常组外,其余各组小鼠均腹腔注射0.12% CCl4油溶液0.1 mL/kg,24 h后断头取血,测ALT、AST,并计算其肝指数.结果 在ANIT所致胆汁瘀积模型中,除三黄颗粒低剂量组AST、ALP差异无统计学意义外(P>0.05),其余对ANIT致小鼠肝指数、ALT、AST、STB、ALP的升高均有抑制作用(P<0.05或P<0.01或P<0.001);在CCl4致小鼠肝损伤模型中,除三黄颗粒低剂量组AST差异无统计学意义(P>0.05)外,其余对CCL4致小鼠肝指数、ALT、AST的升高均有抑制作用(P<0.05或P<0.01或P<0.001).结论 三黄颗粒的退黄作用与其保肝利胆作用有关.

Abstract

Objective To study the mechanism of receding jaundice of sanhuang granule primitively. Methods The cholestasis model induced by α-naphthy-isothiocyanate ( ANTT) was established in mice. The mice were randomly divided into 6 groups: control group, ANIT-injured group, yinzhihuang granule group, low dose, medium dose, high dose of sanhuang granule. Medicine was given by intragatrical injection( ig) for 10 d,70 mg/kg ANIT was given by ig except control group on 8 th day,then medicine was given continuously. Blood samples were taken by cutting heads 48 h after poisoning. Alanine aminotransferase( ALT) , aspartate aminotransferase( AST) , serum total bilirubin ( STB) ,and alkaline phosphatase( ALP) were determined,livers were cut out for liver index;The liver-injury model induced by carbon tetrachloride ( CCl4 ) was established in mice. The mice were randomly divided into 7 groups: control group, CCl4 -injured group, bifendate pills group, yinzhihuang granule group, low dose, medium dose, high dose of sanhuang granule. Medicine was given by ig for 10 d,0. 12% CCl4 was given by ip 2 h after the last medication except control group. Blood samples were taken by cutting heads 24 h after poisoning. ALT, AST were determined, livers were cut out for liver index. Results In the cholestasis model induced by ANIT,the increases of liver index, ALT,AST,STB, ALP were inhibited bbviously by sanhuang granule, but the low dose had no effect on AST and ALP. In the liver-injury model induced by CCl4 ,the increases of liver index, ALT, AST were prevented significantly by sanhuang granule,but the low dose had no effect on AST. Conclusion The mechanism of receding jaundice of sanhuang granule is related to its liver protection and cholagogue function.
