Effects and Mechanism of Gregen(Puerariae Lobatae Radix)Combined with Chitosan on Lipid Metabolism in Obese Mice Induced by High Fat Diet
Objective To explore the effects of Gregen(Puerariae Lobatae Radix)ethanol extract combined with chitosan on lipid metabolism in obese mice.Methods The obesity model of C57BL/6J mice was established by high-fat diet induction.At the same time,Gregen(Puerariae Lobatae Radix)ethanol extract combined with chitosan was given to intervene for 7 weeks.Food intake,body weight changes and body fat of mice in each group were compared,and serum lipid levels,epididymal fat pad and liver fat weight.Pathological changes of liver and kidney lesions were observed by HE staining.Western blot was used to detect the HMGCR,TNF-α and NF-κB protein expressions in liver tissue.Results There was no significant difference in food intake among all groups.Compared with the blank control group,the high-fat diet mice showed a significant increase in body weight,abdominal obesity,increased body fat content and liver fat deposition.The weight gain of pueraria combined with chitosan group was slow and the body fat was normal.The levels of total triglyceride,total cholesterol and low density lipoprotein cholesterol in serum were significantly lower than those in the model group,while the levels of high density lipoprotein cholesterol were significantly increased.No fat infiltration was observed in the liver of mice in the pueraria combined with chitosan group,and the liver fat infiltration was alleviated in the chitosan and pueraria groups,while obvious fat infiltration was observed in the model group.The expression levels of HMGCR,TNF-α and NF-κB in liver tissue were decreased(P<0.05).Conclusion Gregen(Puerariae Lobatae Radix)ethanol extract combined with chitosan can improve the lipid metabolism disorder of obese mice,reduce the level of serum invertase and inflammatory factors,and have the potential function of regulating body fat.
