Experimental Study on Dabuyuanjian(大补元煎)Increasing the Sensitivity of Esophageal Cancer Cells to Platinum Drugs through MAPK Pathway
Objective The purpose of this study is to comprehensively analyze BATMAN-TCM database,TCGA database,cell biology verification and tumor-bearing experiments in nude mice,and to deeply explore the potential mechanism of Dabuyuanjian(大补元煎)in the treatment of esophageal cancer,especially its influence on the sensitivity of cisplatin treatment.Methods Firstly,the composition of Dabuyuanjian and its effect on target protein were comprehensively analyzed by BATMAN-TCM database,and the correlation enrichment analysis was carried out.Secondly,the differential genes of esophageal cancer were screened in TCGA database and analyzed in detail.Subsequently,the sensitivity of Dabuyuanjian to cisplatin in esophageal cancer cells was verified by cell biology.Finally,the experiment of nude mice bearing tumor proves that Dabuyuanjian can enhance the inhibitory effect of cisplatin on tumor.Results The analysis results of BATMAN-TCM and TCGA showed that the target protein of Dabuyuanjian played an important role in cell response to chemical stimulation,metabolic pathway,cancer pathway and other key functions.Experimental verification shows that Dabuyuanjian can not only significantly enhance the sensitivity of esophageal cancer cells to cisplatin,but also further enhance the therapeutic effect of cisplatin by activating MAPK pathway.Cell proliferation and tumor-bearing experiments in nude mice confirmed the synergistic effect of Dabuyuanjian and cisplatin,which significantly inhibited the proliferation of esophageal cancer cells and the growth of tumor-bearing mice.Conclusion This study concludes that Dabuyuanjian may improve the sensitivity of cisplatin to esophageal cancer by activating MAPK pathway,which provides a new theoretical and experimental basis for the application of Dabuyuanjian in cancer treatment.
