萆薢渗湿汤对急性湿疹小鼠模型的作用机制研究

目的 探讨萆薢渗湿汤对急性湿疹小鼠模型的治疗效果及作用机制。方法 选用SPF级KM雄性小鼠60只,适应性喂养7 d后随机分成6组,每组10只,采用涂抹2,4-二硝基氯苯丙酮溶液的方法进行造模,分为正常对照组、模型对照组、强的松阳性药物组[药物剂量为10 mg/(kg·d),相当于临床剂量的10倍]、萆薢 渗湿汤低、中、高剂量组(药物剂量分别为0。70、1。40、2。80 g/kg,相当于临床等效剂量的0。5、1、2倍)。模型激发后24 h,强的松阳性药物组,萆薢渗湿汤高、中、低剂量组小鼠每日分别通过灌胃给予相应药物0。1 mL/10 g,连续给药7 d,给予正常对照组和模型对照组纯化水,在此期间正常喂养。观察各组小鼠的一般情况、耳朵皮损的改善情况、脾脏指数并采用ELISA法检测各组小鼠体内肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-2(interleukin-2,IL-2)、白细胞介素-4(interleukin-4,IL-4)。结果 与正常对照组比较,模型对照组小鼠右耳重量、厚度均显著增加(P<0。01,P<0。01),左耳厚度、重量变化不显著;与模型对照组比较,强的松阳性药物组与萆薢渗湿汤高中剂量组的右耳重量显著减轻(P<0。01),强的松阳性药物组和萆薢渗湿汤高剂量组的右耳厚度显著降低(P<0。01),强的松阳性药物组和萆薢渗湿汤各剂量组左耳重量、厚度变化均不显著。与正常对照组比较,模型对照组小鼠脾脏指数显著升高(P<0。01);与模型对照组比较,强的松阳性药物组与萆薢渗湿汤高剂量组脾脏指数显著降低(P<0。05,P<0。01)。与正常对照组比较,模型对照组小鼠血清中IL-2、IL-4、TNF-α含量均显著升高(P<0。01);与模型对照组比较,强的松阳性药物组和萆薢渗湿汤各剂量组小鼠血清中IL-2、IL-4、TNF-α含量显著下降(P<0。01)。结论 萆薢渗湿汤可以治疗小鼠急性湿疹模型的皮肤炎症,其机制可能与降低小鼠血清中炎症因子L-2、IL-4、TNF-α的含量有关。
Study on the Mechanism of Action of Bixie Shenshi Decoction(萆薢渗湿汤)on Acute Eczema Mouse Model
Objective To explore the therapeutic effect and mechanism of Bixie Shenshi Decoction(萆薢渗湿汤)on acute eczema mouse model.Methods Sixty SPF KM male mice were randomly divided into 6 groups after adaptive feeding for 7 days,with 10 mice in each group.The model was established by applying 2,4-dinitrochlorophenylpropanone solution.The rats were divided into normal control group,model control group,prednisone positive drug group[drug dose of 10 mg/(kg·d),equivalent to 10 times the clinical dose],Bixie Shenshi Decoction low,medium and high dose group(drug doses were 0.70,1.40,2.80 g/kg,equivalent to 0.5,1,2 times the clinical equivalent dose).Twenty-four hours after the model was stimulated,the mice in the prednisone positive drug group and the high,medium and low dose groups of Bixie Shenshi Decoction were given 0.1 mL/10 g of the corresponding drugs by gavage once a day for 7 days.The normal control group and the model control group were given purified water,and the mice were fed normally during the administration.The general condition of the mice in each group,the improvement of ear skin lesions,and the spleen index were observed.The tumor necrosis factor-α(TNF-α),interleukin-2(IL-2),and interleukin-4(IL-4)in each group were detected by ELISA.Results Compared with the normal control group,the weight and thickness of the right ear of the mice in the model control group increased significantly(P<0.01),and the thickness and weight of the left ear did not change significantly.Compared with the model control group,the weight of the right ear in the prednisone positive drug group and the high and middle dose of Bixie Shenshi Decoction groups were significantly reduced(P<0.01),and the thickness of the right ear in the prednisone positive drug group and the high dose of Bixie Shenshi Decoction group were significantly reduced(P<0.01).There was no significant change in the weight and thickness of the left ear in the prednisone positive drug group and each dose of Bixie ShenShi Dection group.Compared with the normal control group,the spleen index of the model control group was significantly increased(P<0.01).Compared with the model control group,the spleen index of the prednisone positive drug group and the high dose of Bixie ShenShi Dection group were significantly decreased(P<0.05,P<0.01).Compared with the normal control group,the serum levels of IL-2,IL-4 and TNF-α in the model control group were significantly increased(P<0.01).Compared with the model control group,the serum levels of IL-2,IL-4 and TNF-α in the prednisone positive drug group and each dose group of Bixie Shenshi Decoction were significantly decreased(P<0.01).Conclusion Bixie Shenshi Decoction can treat skin inflammation in acute eczema model of mice,and its mechanism may be related to the reduction of inflammatory factors L-2,IL-4 and TNF-α in serum of mice.
