Study on Quality Evaluation and Determination of Main Components of Digupi(Lycii Cortex)by Fingerprints and Chemometrics Methods
Objective To establish the fingerprints of Digupi(Lycii Cortex)from eleven producing areas,and to determine the contents of lycii B,lycii A,scopolin and cinnamic acid.Methods A Kromasil 100-5C18(4.6 mm × 250 mm,5 µ m)column was used as the chromatographic column for this experiment;the mobile phase was acetonitrile(A)-0.3%aqueous trifluoroacetic acid(B)for gradient elution;the flow rate was 1.0 mL/min;the ultraviolet(UV)detection wavelength was 278 nm;the column temperature was 30 ℃;and the injection volume was 10 μL.Results The content ranges of ethyl dermatophyllin(1.547 5±1.481 7)%,methyldermatophyllin(0.373 7±0.306 9)%,scopolamine(0.029 1±0.024 7)%,and the content ranges of cinnamic acid(0.029 1±0.024 7)%were not marked because of low cinnamic acid content and cinnamic acid was not detected in some of the origins.A total of 8 common peaks were identified in the eleven habitats,and the similarity was above 0.895.After that,cluster analysis,principal component analysis and partial least squares discriminant analysis were carried out on the 20 batches of Digupi(Lycii Cortex)by the peak area of the common peaks.Conclusion The fingerprint analysis method of Digupi(Lycii Cortex)crataegi established in this experiment has high credibility,and the methodological investigation is in accordance with the regulations.Through the combination of fingerprinting and content determination,the chemometric analysis method was applied to establish the quality evaluation model of Digupi(Lycii Cortex),which made a certain contribution to better promote the application of Digupi(Lycii Cortex)in the clinical aspects of traditional Chinese medicine.
