Optimization of Extraction Technology and Antioxidant Activity of Diphenylheptane from Gaoliangjiang(Alpinia Officinarum Rhizoma)by Response Surface Methodology
Objective To optimize of extraction process of diphenylheptane from Gaoliangjiang(Alpinia Officinarum Rhizoma)by response surface methodology and study its antioxidant activity in vitro experiments.Methods The diphenylheptane of Gaoliangjiang(Alpinia Officinarum Rhizoma)was extracted by cold soaking with dichloromethane.With extraction times,extraction time and solid-liquid ratio as the variables,The extraction rate of diphenylheptane of Gaoliangjiang(Alpinia Officinarum Rhizoma)as the index,Box-Behnken response surface methodology was used to analyze the influence of various factors on extraction rate and determine the optimal extraction conditions.The antioxidant capacity of diphenylheptane from Gaoliangjiang(Alpinia Officinarum Rhizoma)was evaluated by Fe3+reduction capacity and the total antioxidant capacity(ABTS method)Results The optimal extraction process:extraction time of 56 hours,solid-liquid ratio of 1∶2,extraction frequency of 2 times.The extraction rate of diphenylheptane was 2.883%with RSD of 1.05%.When the concentration of diphenylheptane extract from Gaoliangjiang(Alpinia Officinarum Rhizoma)was 0.25 mg/mL,the maximum value of Fe3+reduction ability and total antioxidant capacity(ABTS method)were 2.49 mmol·g-1 and 2.10 mmol·g-1,respectively.Conclusion The optimized extraction process of diphenylheptane from Gaoliangjiang(Alpinia Officinarum Rhizoma)is stable and feasible and simple.The diphenylheptane extract of Gaoliangjiang(Alpinia Officinarum Rhizoma)has good antioxidant capacity in vitro.
