Determination of dhurrin content in brewing sorghum from different producing regions by high performance liquid chromatography-tandem mass spectrometry
A high efficiency,accurate,simple,and convenient method to detect the content of dhurrin in brewing sorghum by high performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS).The sample was extracted by 95%methanol,separated by UPLC BEH C18 column,and then detected by HPLC-MS/MS under multiple reaction monitoring mode(MRM).The results showed that the limits of detection(LOD)of dhurrin was 1.0 ng/mL,while the limit of quantification(LOQ)was 3.0 ng/mL,and a good linear relationship was shown in the range of 5.0 to 200.0 ng/mL,with a correlation coefficients(R2)of 0.9991526,the relative standard deviations(RSD)was 2.29%,the recovery rate for three levels of spiking ranges from 94.3%to 101.5%Among the six main producing provinces of sorghum in Heilongjiang,Jilin,Liaoning,Shaanxi,Shanxi,and Guizhou,the dhurrin content in brewing sorghum from Shaanxi Province was the lowest(1.45 mg/kg),and the highest content(3.50 mg/kg)was detected in brewing sorghum from Jilin Province.Among three producing regions of sorghum in Southwest,North China,and Northeast China,the dhurrin content in brewing sorghum from Southwest China was the lowest(1.84 mg/kg),and the highest content(3.97 mg/kg)was detected in brewing sorghum from North China.The content of dhurrin in two high-quality brewing sorghum varieties,Hongyingzi and Jinza-22,was low.The study indicated con-tent of dhurrin in brewing sorghum varied greatly from different producing regions and varieties with high-quality sorghum having a lower content of dhurrin.
