Detection of Soybean Isoflavone Content by Ultra-High Performance Liquid Chromatography
Objective:To establish an efficient detection method for determining the content of soy isoflavones in soybeans and study the differences in soy isoflavone components.Method:Ultra high performance liquid chromatography was used to detect the content of soy isoflavones in soybeans.A 70%methanol solution was added to the soy sample,and soy isoflavones were extracted by ultrasonic method.The chromatographic column was BEHC C18 column(150 mm×2.1 mm,1.7 μm),the mobile phase is acetonitrile-0.2%acetic acid,the detection wavelength is 260 nm,the column temperature is 30℃,and the flow rate is 0.15 mL·min-1.Result:The linear relationship,precision,and stability of the four soybean isoflavones were good,with an average recovery rate of 97.6%to 98.6%.The content of daidzin,the isoflavone component of soybeans,is the highest,while the content of daidzein is the lowest.The specific content is daidzin 352.0 μg·g-1,genistein 88.0 μg·g-1,genistein 25.8 μg·g-1,daidzein 19.5 μg·g-1.Conclusion:Ultra high performance liquid chromatography has the advantages of simple operation,fast detection speed,and high sensitivity,and can be used for the detection of soy isoflavone content in soybeans.
