高效液相色谱法测定食品中的抗坏血酸2-葡萄糖苷

摘要
建立了高效液相色谱法测定食品中维生素C衍生物抗坏血酸 2-葡萄糖苷含量的方法.试样经偏磷酸溶解超声提取后,经Zorbax SB-C18(4.6 mm×250 mm,5 μm)色谱柱分离,高效液相色谱紫外检测器检测,根据保留时间定性,用外标法定量.结果显示,该方法的线性关系良好,线性相关系数为 1.000,方法精密度为0.032%(n=6),重复性为 0.023%(n=6),回收率在 97.2%~98.6%,相对标准偏差≤2.13%(n=6).该方法前处理简单,回收率高,重复性好,适用于食品中抗坏血酸 2-葡萄糖苷的测定.
Abstract
A method for the determination of ascorbic acid 2-glucoside,a derivative of vitamin C,in food by high performance liquid chromatography was established.The samples were ultrasonically extracted with metaphosphoric acid,separated on a Zorbax SB-C18(4.6 mm×250 mm,5 μm)column and detected by high performance liquid chromatography with ultraviolet detector.Qualitative analysis was based on retention time and quantitative analysis was based on external standard method.The results showed that the linear relationship of the method was good,the linear correlation coefficient was 1.000,the precision of the method was 0.032%(n=6),the repeatability was 0.023%(n=6),the recovery rate was 97.2%~98.6%,and the relative standard deviation was≤2.13%(n=6).The method is simple in pretreatment,high in recovery,good in reproducibility and suitable for the determination of ascorbic acid 2-glucoside in food.
