Study on Optimum Conditions for Determination of Protein in Pleurotus eryngii by Kjeldahl Nitrogen Determination Method
The Kjeldahl nitrogen determination method was used to detect the protein content of Pleurotus eryngii,and the digestion time,digestion temperature and catalyst dosage were optimized by single factor and response surface tests.The results showed that the optimal parameters for the detection of protein content in Pleurotus eryngii by Kjeldahl nitrogen determination method were the digestion time of 1.85 h,the digestion temperature of 438℃,and the amount of concentrated sulfuric acid in the catalyst was 14.8 mL,and the protein content of Pleurotus eryngii was the highest(20.71 g/100 g)under these conditions.Compared with the national standard method,the optimized method in this study improved the detection efficiency of protein content,reduced the amount of concentrated sulfuric acid in the catalyst,and shortened the digestion time.
