为优化动物双歧杆菌乳亚种真空冷冻干燥保护剂的配方,通过单因素试验并结合SPSS软件统计分析,得出动物双歧杆菌乳亚种冷冻干燥保护剂中对存活率有较大影响的因素;以存活率为指标,选择单因素试验结果中对存活率影响较大的 3 个因素进一步进行响应面试验优化,确定动物双歧杆菌乳亚种冷冻干燥保护剂最佳配方.以冻干后动物双歧杆菌乳亚种菌株存活率为响应值,利用响应面分析法优化的结果为脱脂奶粉11.31 g/100 g、海藻糖 13.96 g/100 g、甘油 1.69 g/100 g,在此条件下,菌体存活率可达 93.31%,该冷冻干燥保护剂配方具有较高的实用价值.
Optimization of Protective Agent Formula for Freeze-drying of Lyophilized Bifidobacterium animalis subsp.lactis by Response Surface Methodology
To optimize the formulation of cryoprotectants for lyophilized Bifidobacterium animalis subsp.lactis,the factors of lyoprotectant that have a greater impact on the survival rate of Bifidobacterium animalis subsp.lactis were obtained through the single factor test combined with the statistical analysis of SPSS software.To optimize the formula of lyoprotectant,three factors that have influence on the survival rate in a single factor experiment were selected for response surface analysis based on the survival rate.The research concluded the optimum ofcryoprotectants as follows:The skimmed milk 11.31 g/100 g,trehalose 13.96 g/100 g,glycerol 1.69 g/100 g.By using this process,the optimal cell survival rate was observed to be 93.31%,it had the features of practicability.
