摘要

目的:研究沙棘果皮中总黄酮的提取与纯化工艺,并评估其抗氧化活性.方法:采用超声波辅助提取沙棘果皮中的黄酮物质,并通过单因素和正交试验优化黄酮提取条件.粗黄酮经X-5型大孔吸附树脂纯化,对比纯化后黄酮和L-抗坏血酸对DPPH自由基和ABTS自由基的清除能力,评价其抗氧化活性.结果:最佳提取条件为料液比1∶30(g∶mL)、乙醇体积分数50%、提取时间70 min,此条件下黄酮提取率达3.62%.经大孔树脂纯化后,黄酮纯度由15.5%提高至49.0%,沙棘果皮总黄酮质量浓度为1.0 mg·mL-1,DPPH自由基清除率为80.37%,ABTS自由基清除率为77.65%.结论:纯化后的沙棘果皮黄酮具有一定的抗氧化活性,可用于天然抗氧剂的开发.

Abstract

Objective:To explore the extraction and purification process of total flavonoids from sea buckthorn peel and evaluate its antioxidant activity.Method:Ultrasonic-assisted extraction of total flavonoids from sea buckthorn peel was carried out,and the extraction conditions of flavonoids were optimized by single factor and orthogonal experiments.Crude flavonoids were purified by X-5 macroporous resin,and the antioxidant activities of flavonoids and ascorbic acid were evaluated by comparing their DPPH free radicals and ABTS free radicals scavenging ability.Result:The optimal extraction conditions were solid-liquid ratio 1∶30(g∶mL),ethanol concentration of 50%,extraction time of 70 min,and under these conditions,the extraction yield of flavonoids reached 3.62%.After purification by macroporous resin,flavonoid purity increased from 15.5%to 49.0%,total flavonoid concentration was 1.0 mg·mL-1,and DPPH free radicals clearance rate was 80.37%,ABTS free radicals clearance rate was 77.65%.Conclusion:The purified flavonoids from sea buckthorn fruit peel have certain antioxidant activity and can be used in the development of natural antioxidant.
